摘  要:目的为研究乌鲁木齐市常见库蚊的种型,利用其形态特征结合分子特征对蚊虫进行快速的种类鉴定。方法研究样本为乌鲁木齐市区采集、人工繁殖的成蚊和幼虫,形态学鉴定并测定试验蚊虫的mtDNA-COI和rDNA-ITS2基因序列进行分子生物学鉴定分析。结果对成蚊及幼虫进行形态鉴定,根据镜下特征判断为尖音库蚊复合组。试验蚊虫mtDNA-COI和rDNA-ITS2的PCR扩增产物测序,对获得的序列进行BLAST同源性比对,结合形态学鉴定等因素确定为尖音库蚊。结论应用形态特征结合分子特征鉴定蚊虫种型具有简单、快速的特性。确认了乌鲁木齐市一种常见蚊种为尖音库蚊,引入了分子生物学鉴定蚊种的方法。Objective To study the categories of common Culex species in Urumchi city , the use of morphologi- cal and molecular characteristics of the mosquito for rapid species identification. Methods The samples collected from the city of Urumqi, the artificial breeding of adult mosquitoes and larvae, morphological identifica- tion and determination of test mosquitog mtDNA -COI and rDNA - ITS2 gene sequences for identification of molecular biology analysis. Results Conduct morphological identification for adult mosquitoes and larvae, to determine the microscopic characteristics of Culex pipiens complex group. Test mosquito mtDNA - COl and rDNA- ITS2 PCR amplification products were sequenced to obtain the sequence BLAST homology comparison, combined with the morphological identification of other factors identified as Culex pipiens. Conclusions Application of morphological and molecular characterization of mosquito species is simple, fast. Urumqi identi- fied a common mosquito species Culex pipiens, introduced the mosquito species identification of molecular biology methods.
