摘  要:目的观察积雪草甙对系统性硬皮病(SSc)患者皮肤成纤维细胞增殖、胶原蛋白合成及分泌转化生长因子β1(TGF-β1)的影响。方法原代培养SSc患者皮肤成纤维细胞,使用不同浓度的积雪草甙作用48h;MTT法、羟脯氨酸法及ELISA法测定积雪草甙对成纤维细胞增殖、胶原蛋白合成及分泌TGF-β1的影响。结果与空白对照组相比,积雪草甙组能抑制成纤维细胞增殖,减少成纤维细胞胶原蛋白合成及TGF-β1的分泌,其作用具有浓度依赖性(P<0.05)。结论积雪草甙治疗SSc的机理与其抑制成纤维细胞增殖、胶原蛋白合成及TGF-β1的分泌有关。Objective To study the effects of asiaticoside on the proliferation, collagen synthesis and transfer growth factor-β1 (TGF-β1) secretion of dermal fibroblasts in the patients with systemic sclerosis(SSc). Methods Dermal fibroblasts from patients with SSc were primary cultured and treated with different concentrations of asiaticoside for 48 hours. The cell proliferation was determined by MTT assay. The collagen production was determined by calculating hydroproline in cultured medium. The level of TGF-β1 in cell culture supernatant was measured with ELISA. Results Compared with blank control group, the proliferation of cultured SSc dermal fibroblasts was inhibited, and collagen synthesis and secretion of TGF-β1 from fibroblasts were reduced in a dose-dependent manner in hydroproline-treated groups (P〈0. 05). Conclusion The mechanism for asiaticoside to treat the patients with SSc is associated with the inhibition of dermal fibroblasts proliferation, collagen synthesis and TGF-β1 secretion.
