摘  要:目的观察右美托咪定对小鼠利多卡因或士的宁所致惊厥作用的影响。方法 64只小鼠均分为八组,分别于腹腔注射不同剂量(0、7.5、15.0和30.0μg/kg)右美托咪定5min后,腹腔注射利多卡因或士的宁建立两种小鼠惊厥模型,观察小鼠惊厥潜伏期、持续期、惊厥数及死亡数;以序贯法测定预先给予右美托咪定5min后利多卡因或士的宁致惊厥的半数有效剂量(ED50)。结果右美托咪定15.0μg/kg和30.0μg/kg缩短利多卡因或士的宁致小鼠惊厥的持续时间(P<0.01)。右美托咪定15.0μg/kg增加利多卡因或士的宁致惊厥的ED50(P<0.05),延长士的宁致惊厥的潜伏期(P<0.01)并减少惊厥数(P<0.05)。结论小鼠腹腔注射右美托咪定15-30μg/kg有抗药物性惊厥的作用。Objective To observe the effects of dexrnedetomidine on convulsion induced by lidocaine or strychnine in mice. Methods A total of 64 mice was equally divided into 8 groups,which were intraperitoneally injected with normal saline and three doses of dexmedetomidine(7.5,15.0 and 30.0μg/kg) at 5 minutes before injecting lidocaine or strychnine to establish two kinds of convulsive mouse models. The latency and duration of convulsion, the incidence of convulsion and death were observed. After intraperitoneal preinjection of dexmedetomidine, the 50% effective dose (ED50) of lidocaine or strychnine for inducing convulsion was determined by sequential method. Results The duration of lidocaine- or strychnine-induced convulsion was shortened by preinjection of dexrnedetomidine 15.0μg/kg and 30.0 μg/kg(P〈0. 01). The ED50 of lidocaine or strychnine was raised(P〈0. 05), the latency period of strychnine-induced convulsion was prolonged(P〈0. 01), and the incidence of strychnine-induced convulsion was reduced by preinjection of dexmedetomidine 15.0 μg/kg(P〈0. 05). Conclusion Intraperitoneal preinjection of dexmedetomidine 15-30 μg/kg can antagonize lidocaine- or strychnine-induced convulsion.
