摘  要:目的 研究吲哚青绿(indocyanine green,ICG)对体外培养的兔晶状体上皮细胞(rabbit lens epithelial cells,RLECs)增殖的影响,探讨其在白内障术中晶状体前囊膜染色技术应用中的安全性、可靠性,并为其是否可在临床推广应用提供理论依据.方法 实验研究.行RLECs体外培养,将不同浓度的ICG加入第二代RLECs中,分别在24 h、48 h、72 h采用MTT比色法及血球板计数法观察不同浓度的ICG对体外培养的RLECs增殖的影响,并在光镜下观察其形态学变化.结果 (1)MTT比色法结果显示:浓度为0.5%和1%的ICG在24 h对RLECs即有明显的促增殖作用,与阴性对照组比较差异具有统计学意义(P<0.05);随作用时间延长,在48、72 h各实验浓度的ICG均对RLECs有明显的促增殖作用,与阴性对照组相比差异具有统计学意义(P<0.05);在同一作用时间内,随ICG浓度增高,对RLECs的促增殖作用逐渐增强,各浓度组间差异具有统计学意义(P<0.05).(2)血细胞板计数法结果:24h血细胞板计数法结果与MTT比色法结果一致.随实验时间的延长,RLECs的黏附性明显增高,无法消化,致使48 h、72 h细胞计数无法进行.结论 ICG能促进体外培养的RLECs的增殖,因此对后囊混浊的形成有促进作用.为减少后囊混浊的发生,白内障手术中应谨慎应用ICG.Objective To study the effects of indocyanine green (ICG) on rabbit lens epithelial cells(RLECs)proliferation by in vitro culture,and to investigate the safety and reliability of the technology of staining the lens capsular with ICG during cataract surgery.Methods RLECs were cultured in vitro.The effects of ICG at different concentrations on proliferation of RLECs in vitro at 24h,48h,72h was measured with MTT staining colorimetric method and cell count technique.The morphological of RLECs changes were observed with light microscope.Results The result of MTT showed that ICG at the concentration of 0.5% and 1% can significantly promote the RLECs proliferation at the time of 24h(P 〈0.05).At 48h and 72h the effects of the proliferation on RLECs of ICG at all of the experimental concentrations were significant (P 〈0.05).At the same time the effects of proliferation were depended on concentrations (P 〈0.05).The result of cell count technique at 24h was consistent to that of MTT.With the prolongation of the time the adhesion of the RLECs was significantly increased,the RLEcs could not be separated,so the cell count could not be implemented at 48h and 72h.Conclusions ICG can promote the RLECs proliferation in vitro,and can contribute to the posterior capsular opacification by this way.In order to decrease the occurrence of PCO,ICG should be carefully used during the cataract surgery.
