摘  要:目的:多糖类产品的质量控制对产品的功效至关重要,本文探讨了黄芪-枸杞复合多糖HPLC特征指纹图谱的构建。方法:首先对复合多糖的降解方法进行了比较,然后对三氟醋酸(TFA)降解法进行了条件优化,在此基础上对10批黄芪-枸杞复合多糖样品进行了比较分析。结果:TFA降解法可以获得较多的黄芪-枸杞复合多糖特征峰,而且当TFA浓度为0.5 mol·L-1、温度为100℃、时间为4 h及存在200r·min-1搅拌时,黄芪-枸杞复合多糖降解组分的色谱特征显著,降解和衍生产物的稳定性以及方法的重现性均达到中药指纹图谱标准。10批复合多糖样品的指纹图谱均具有较强的特征性,整体相似度大于0.9。结论:所建立的"降解-PMP柱前衍生化-HPLC检测"法快速简单、灵敏度高、稳定可靠,可用于复合多糖产品的质量控制。OBJECTIVE To establish a fingerprint profiling method based on HPLC with an attempt for quality control of polysaccharides. METHODS Complex Astragalus-Lycium polysaecharides were hydrolyzed and derivatized prior to fingerprinting analysis using HPLC. Then the TFA method was further optimized to obtain a combination of reactive conditions for controllable degradation of the polysaccharides, including TFA concentration, temperature, time and agitation. Finally, 10 batches of complex Astragalus-Lycium polysaccharide samples were degraded with the optimized TFA method and analyzed with HPLC. RESULTS Hydrolysis with weak trifluoroacetic acid (TFA) was found to be appropriate for its simplicity and ef- fectiveness. Under the optimized conditions, the characteristic peaks in the fingerprints of complex polysaccharides were evidently observed with good stability and reproducibility. Furthermore, the similarity of the fingerprints from 10 batches of complex polysaccharides was found to be ≥0. 9. CONCLUSION These results demonstrate the method described here is simple, rapid, sensitive and reliable for fingerprint profiling of the complex Astragalus-Lycium polysaccharides and thus provide a basis for quality control of polysaccharide-containing products.
