摘 要:目的探讨橙皮素对血管紧张素Ⅱ(AngⅡ)诱导H9C2心肌细胞肥大的影响。方法使用不同浓度橙皮素(0.125、0.25、0.5、1μmol/L)和AngⅡ(1μmol/L)共同孵育H9C2心肌细胞12h,心肌细胞骨架骨骼α肌动蛋白(α-actinin)荧光染色评估H9C2细胞面积改变以及real-time PCR方法检测心肌肥厚标志物BNP、β-MHC的mRNA表达变化,以观察不同浓度橙皮素对AngⅡ诱导H9C2心肌细胞肥大的影响,选择0.25μmol/L橙皮素和AngⅡ(1μmol/L)共同孵育H9C2心肌细胞6、12、24h,观察橙皮素对AngⅡ诱导H9C2心肌细胞肥大抑制作用的时间相关性。结果橙皮素干预可以缓解AngⅡ诱导H9C2心肌细胞的细胞面积增大;橙皮素抑制了AngⅡ诱导的H9C2心肌细胞BNP、β-MHC的mRNA表达水平升高。结论橙皮素能够抑制AngⅡ诱导H9C2心肌细胞肥大,其有可能成为治疗心肌重构新的潜在药物。Objective To investigate the effect of hesperetin on H9C2 cardiomyocytes hypertrophy induced by Ang Ⅱ. Methods The embryonic rat heart - drived H9C2 cells were coincubated with different concentrations of hesperetin(0. 125, 0.25, 0.5 and 1 μmol/L) and Ang Ⅱ (ltxmolfL) for 12h. We characterized cells by immunocyto -chemistry for cardiac α -actinin to examine increasing cell surface area and real - time PCR was used to measure mRNA levels of hypertrophic markers BNP, and β - MHC to clear the effect of hesperetin on H9C2 cardiomyocytes hypertrophy. Results We found H9C2 cells treated with hesperetin could be protected from Ang Ⅱ - induced hypertrophy. The inceasing mRNA levels of BNP and β- MHC in Aug Ⅱ - stimulated cells were also down - regulated by hesperetin treatment. Conclusion Hesperetin attenuates H9C2 cardiomyocytes hypertrophy which induced by AngⅡ.
