摘  要:目的:观察丹参酮ⅡA磺酸钠(STS)对膀胱出口部分梗阻(PBOO)大鼠膀胱中a-SMA表达的影响,探究其可能的作用机制。方法:通过膀胱颈部分结扎法建立大鼠PBOO模型,24只SD大鼠随机分为PBOO组、STS治疗组(STS组)以及假手术组(Sham组),每组8只。STS组每天给予STS腹腔注射(10 mg/kg),持续4周。PBOO组及Sham组给予等量生理盐水腹腔注射。应用透射电镜及Masson染色检测膀胱组织中胶原纤维的表达,免疫组化和RT-PCR检测膀胱中a-SMA的蛋白及mRNA的表达,应用Western blot检测膀胱组织中ERK1/2、P-ERK1/2蛋白的表达。结果:PBOO组胶原纤维表达量高于Sham组(P<0.05),STS组胶原纤维表达量低于PBOO组(P<0.05)。PBOO组a-SMA蛋白及mRNA表达高于Sham组(P<0.05),STS组低于PBOO组(P<0.05)。三组ERK1/2蛋白表达差异无统计学意义(P>0.05),P-ERK1/2蛋白的表达,PBOO组高于Sham组,STS组低于PBOO组(P<0.05)。结论:STS能够抑制PBOO大鼠a-SMA的表达,其机制可能是通过抑制MAPK通路中ERK1/2蛋白的活化实现的。Objective:To observe the inhibitory effect of sodium tanshinoneⅡ A sulfonate(STS)on a-smooth muscle actin(a-SMA)in partial bladder outlet obstruction(PBOO)rats’ bladder and to explore its possible mechanism.Method:Establishment of PBOO rats models were made by partial ligation of bladder neck.Twenty-four SD rats were randomly divided into PBOO group,STS group and Sham group on average.STS group was given intraperitoneal injection of STS(10mg/kg)for four weeks,and PBOO group and Sham group were given normal saline by intra-peritoneal injection.Collagen fiber of bladder tissue was detected using transmission electron microscope and Masson staining.Then,the expression of a-SMA protein and mRNA were detected by immunohistochemistry and RT-PCR,and expression of ERK1/2,P-ERK1/2protein was explored by Western blot.Result:Collagen expression of PBOO group was higher than that of Sham group,and STS group was lower than PBOO group(P 〈0.05).The expression of a-SMA protein and mRNA of PBOO group were higher than those of Sham group(P 〈0.05),while STS group was lower than PBOO group(P 〈0.05).However,the expression of ERK1/2protein among three groups showed no significant differences(P 〉0.05).The expression of P-ERK1/2protein of PBOO group was higher than that of Sham group,and STS group was lower than PBOO group(P 〈0.05).Conclusion:STS can inhibit the expression of a-SMA protein in PBOO rats,and its mechanism may be related to inhibition of ERK1/2protein activation in MAPK pathway.
