大苞鞘石斛胚组织培养及植株再生研究

摘　　要：为筛选出大苞鞘石斛各生长阶段最适培养基配方,并建立大苞鞘石斛快繁体系,以大苞鞘石斛种胚为材料,以1/2MS为基本培养基,通过正交试验研究不同种类激素和添加物的浓度配比对种胚萌发和植株再生的影响。结果表明,大苞鞘石斛种胚萌发和原球茎膨大的最适培养基为1/2 MS+IBA 1.0 mg·L-1+6-BA 2.0 mg·L-1+NAA 2.0 mg·L-1+香蕉泥50 g·L-1,萌发率达91.29%;叶芽分化的最适培养基为1/2MS+IBA 0.5 mg·L-1+6-BA 1.0 mg·L-1+NAA 0.5 mg·L-1+香蕉泥50 g·L-1;干物质积累的最适培养基为1/2 MS+IBA 2.0 mg·L-1+6-BA 2.0 mg·L-1+NAA 0.5 mg·L-1+香蕉泥200 g·L-1;丛生芽增殖的最佳培养基为1/2 MS+IBA 0.5 mg·L-1+6-BA 1.0 mg·L-1+NAA 0.5 mg·L-1+香蕉泥100 g·L-1,平均增殖系数达6.33;丛生芽生根培养的最佳培养基为1/2 MS+IBA 0.5 mg·L-1+6-BA 3.0 mg·L-1+NAA0.2 mg·L-1+香蕉泥50 g·L-1+土豆泥50 g·L-1。The tissue culture and plant regeneration of Dendrobium wardianum were studied by using embryos as material.The embryos were cultivated in 1/2 MS medium using different plant hormones and additives with different concentrations by orthogonal design test to examine those factors which affect the germination and regeneration, the perfect mediums for different growth periods were screened, and the rapid propagation system of D.wardianum.The results indicated as follows.The optimum medium for the embryo germination and enlargement was 1/2 MS ＋IBA 1.0 mg· L-1 ＋6-BA 1.0 mg· L-1 ＋NAA 2.0 mg· L-1 ＋banana mud 50 g· L-1 with the germination rate of 91.29%.The optimum medium for differentiation was 1/2 MS＋IBA 0.5 mg· L-1 ＋6-BA 1.0 mg· L-1 ＋NAA 0.5 mg· L-1 ＋banana mud 50 g· L-1 .The optimum medium for dry matter accumulation was 1/2 MS＋IBA 2.0 mg· L-1 ＋6-BA 2.0 mg· L-1 ＋NAA 0.5 mg· L-1 ＋banana mud 200 g· L-1 .The optimum medium for proliferation of cluster buds was 1/2 MS ＋IBA 0.5 mg· L-1 ＋6-BA 1.0 mg· L-1 ＋NAA 0.5 mg· L-1 ＋banana mud 100 g· L-1 with the average proliferation rate of 63.3 .The optimum medium for rooting was 1/2MS ＋IBA 0.5 mg· L-1 ＋6-BA 3.0 mg· L-1 ＋NAA 0.2 mg· L-1＋banana mud 50 ·g L-1 ＋mashed potato 50 g· L-1 .
