腰痛宁胶囊及其拆方对Ana-1细胞免疫功能的影响

摘　　要：目的评价腰痛宁全方、拆方及其所含各中药活性部位对小鼠Ana-1细胞免疫调节作用。方法将腰痛宁胶囊中生物碱类、黄酮类、皂苷类、挥发油(含水提液)类、多糖类等活性部位按照腰痛宁组方原则及组方比例,配置了由不同活性部位混合组成的6个腰痛宁组方及拆方样品,用酶联免疫吸附测定(ELISA)方法测定各样品促进Ana-1细胞分泌IL-1β、IL-6、TNF-α因子的EC50。结果腰痛宁全方促进IL-1β分泌的EC50最低,不含药引黄酒多糖的腰痛宁活性部位组方次之,其他样品无促进IL-1β分泌的EC50;6个样品均有促进IL-6和TNF-α分泌的活性且生物碱+黄酮+皂苷组方的EC50最低,但这两个指标下该样品与腰痛宁全方的EC50无显著性差异(α=0.05)。结论 6个样品中腰痛宁全方在3个评价指标下均表现出良好的调节机体免疫的作用,显示出腰痛宁全方的合理性与增强免疫调节的优势。AIM To evaluate the effect of active fractions of the material medicines used in Yaotongning Capsules whole formula and decomposed formulas of Yaotongning on immune function of Ana-1 cells of mice.METHODS The whole and five decomposed formulas were prepared by mixing different active fractions including alkaloids,flavonoids,saponins,volatile oil/water extracts and polysaccharides according to the ratio of materials Yaotongning and formulating principle of Yaotongning.The Enzyme-Linked Immuno Sorbent Assay （ELISA） was used to detect the EC50 of the samples＇ promoting the secretion of IL-1β,IL-6 and TNF-α.RESULTS The whole formula of Yaotongning had the best effect on promoting IL-1β secretion,and whole formula without rice polysaccharides took the second place while the other four samples had no EC50 of promoting IL-1β secretion.All six samples had the effect on promoting the secretion of IL-6 and TNF-α.The combination of alkaloids,flavonoids and saponins of Yaotongning had the minimum EC50 but there was no significant difference （α =0.05） between this combination and the whole formula of Yaotongning on above two cytokines.CONCLUSION Among the six combinations,the whole formula of Yaotongning consisting of all active fractions in Yaotongning exhibits good immunomodulatory effect under the three evaluating indicators,showing the reasonability and advantage of Yaotongning Capsules＇ prescription on enhancing immune regulation.
