雄黄、冰片和珍珠粉对人牙周膜细胞增殖和碱性磷酸酶活性的影响

摘  要:目的:探讨雄黄、冰片和珍珠粉按1∶3∶1的重量比配伍组成的自制复方中药制剂对人牙周膜细胞(periodontal ligament cell,PDLC)增殖和碱性磷酸酶(alkaline phosphatase,ALP)活性的影响。方法:将5种不同浓度的复方中药制剂作用于体外培养的PDLC。MTT法观察PDLC的增殖情况,酶动力学方法检测PDLC的碱性磷酸酶活性。氢氧化钙溶液作为对照组。结果:将浓度在0.00125%-0.02%范围内的复方中药制剂分别作用于PDLC24、48和72h后,发现PDLC的增殖和碱性磷酸酶活性随药物浓度和作用时间的增加而降低,抑制作用最大的浓度和作用时间分别为0.02%和72h。结论:自制复方中药制剂对PDLC的增殖和碱性磷酸酶活性具有一定的抑制作用。Objective:To examine the effect of the compound traditional Chinese medicine made from realgar,borneol and pearl powder on the proliferation and the alkaline phosphatase(ALP)activity of the periodontal ligament cell(PDLC).The proportion by weight of borneol,realgar and pearl powder was 3:1:1.Methods:We have added five different concentrations of the compound traditional Chinese medicine on the PDLC in vitro.Then we have observed the proliferation of PDLC by MTT and detected ALP activity of PDLC by enzyme dynamics.The same concentration of calcium hydroxide has been chosen as a contrast.Results:The compound traditional Chinese medicine could significantly inhibit the proliferation and ALP activity of PDLC which were positively correlated to the concentration and the reaction time at the concentration in the range of 0.00125%to 0.02% within 72 hours after reaction.The maximum inhibitory concentration and reaction time were respectively 0.02% and 72 hours.Conclusion:The compound traditional Chinese medicine made from realgar,borneol,and pearl powder has a certain inhibitory effect on the proliferation and ALP activity of PDLC.
