摘 要:目的:探讨蒺藜皂苷对大鼠缺血再灌注损伤后脑组织的保护作用及其机制。方法:将SD大鼠随机分为假手术组、模型组和蒺藜皂苷治疗低剂量组和高剂量组,采用线栓法制备大鼠脑缺血再灌注损伤模型。脑缺血再灌注24h后检测大鼠神经功能损伤评分;干湿重法测定脑含水量;ELISA法检测大鼠脑组织及血清TNF-α、IL-6和IL-10的含量。结果:与模型组相比较,蒺藜皂苷治疗组可明显减轻脑缺血再灌注大鼠神经功能损伤和脑水肿程度,降低缺血脑组织和血清TNF-α和IL-6的含量、增加缺血脑组织和血清IL-10的含量。结论:蒺藜皂苷对大鼠脑缺血再灌注损伤具有保护作用,其机制可能与其增强IL-10的表达,降低TNF-α和IL-6的表达,从而抑制炎症反应、减轻大脑神经功能损伤有关。Objective:To study the neuroprotective effect of gross saponins of tribulus terrestris on the cerebral tissues after ischemia-reperfusion injury and its mechanisms in rats.Methods :The SD rats were divided into sham operation group, cerebral ischemia-reperfusion model group, gross saponins of tribulus terrestris low-dose group and high-dose group.Cerebral ischemia reperfusion model was established with suture emboli method in middle cerebral artery of rats.Neurological deficit scores were measured 24 hours after the cerebral reperfusion.The water content was calculated with dry and wet weight method; Content of tumor necrosis factor-α(TNF-α), interleukin-6 (IL-6) and interleukin-10 (IL-10) in rat brain and in serum was detected by ELISA.Results:Neu-rological deficit scores, the brain water content and expression levels of TNF-αand IL-6 in the cerebral tissues and in serum were significantly lower in gross saponins of tribulus terrestris treatment groups than ischemia-reperfusion model group.However, gross saponins of tribulus terrestris could significantly increased IL-10 expression level.Conclusion: Gross saponins of tribulus terrestris exerts the neuroprotective effect on cerebral ischemia-reperfusion injury in rats through inhibiting the inflammatory reaction, which may be associated with the increase of the IL-10 expression and decrease of the TNF-αand IL-6 expression.
