摘  要:目的提高金钗石斛种子的萌发率,探索诱导愈伤组织的培养基。方法以金钗石斛种子为材料,研究其种子的萌发和愈伤组织的诱导条件。结果金钗石斛种子在MS+土豆块10 g/L+琼脂7 g/L+蔗糖30 g/L的培养基中萌发最快,时间为9 d,萌发率最高达92.5%;培养基(MS+0.2 g/L碳颗粒+琼脂7 g/L+蔗糖30 g/L)能够诱导金钗石斛原球茎形成愈伤组织。结论在MS培养基中单独添加土豆时能提高金钗石斛成熟种子的萌发率;碳粉能有效促进金钗石斛愈伤组织的形成。Objective To promote the germination rate and to obtain the callus of Dendrobium nobile Lindle, we explored the media components through tissue culturing. Methods The seeds of D. nobile were applied to observe the germination condition and the callus induction in tissue culture. Results High germination rate (92.50%) was screened from the culture medium with the shortest time of 9 days, i.e. Murashige and Skoog medium (MS) + 10 g/L potato pieces + 7 g/L agar + 30 g/L sucrose. Moreover, the callus was induced from the optimized culture medium as MS + 0.2 g/L carbon powder + agar 7 g/L + 30 g/L sucrose. Conclusion The germination rate could he significantly promoted in MS medium combination with potato. And carbon powder probably improves the callus induction in D. nobile.
