摘 要:目的 观察血液血小板膜糖蛋白Ⅰbα(GPⅠbα)、血管性血友病因子(vWF)表达水平与静脉血栓性疾病(VTE)进展的相关性。方法 SD大鼠下腔静脉(IVC)血栓造模,检测造模后2、8、24、72 h各组大鼠血液中GPⅠbα、vWF的变化情况;收集30例DVT患者、30例实验对照患者、30例健康对照者血样,ELISA法检测GPⅠbα、vWF的表达水平并进行比较分析。结果 大鼠造模后,假手术组与模型组血GPⅠbα、vWF水平均呈增高趋势。在下腔静脉受损、血栓形成的起始阶段(2 h),GPⅠbα、vWF水平未明显增高;而在2~24 h血栓形成高峰阶段,假手术组与模型组GPⅠbα、vWF水平升高,模型组GPⅠbα、vWF升高更显著;24~72 h稳定血栓形成,GPⅠbα水平降低,模型组和假手术组vWF水平仍表现高于2 h和对照组。DVT患者组GPⅠbα、vWF表达水平高于无DVT患者组及健康人对照组(P〈0.05)。结论 血GPⅠbα、vWF表达水平随DVT发生而升高,与VTE的进展相关。Objective To observe the association between platelet membrane glycoproteinIbα (GP I bα) ,von willebrand factor (vWF) and disease development of venous thromboembolic events(VTE). Methods The animal models of inferior vena tara(IVC) thrombus was established with SD rats and the expressions of GPIbα and vWF in rat blood at 2, 8, 24 and 72 hour after modeling were determined by ELISA. The blood samples of 30 deep venous thrombosis(DVT) patients were collected as the experimental group and the levels of GPIbα and vWF were compared with those of 30 non-DVT patients and 30 healthy subjects. Results The expressions of GPIbα and vWF in the rat blood of modeling group and Sham operation group showed significantly up-regulated( P 〈 0.05 ) in 8 and 24 hour. The levels of GPIbα and vWF in DVT patients and non-DVT patients were higher than those of healthy subjects, and DVT patients were higher than non-DVT patients ( P 〈 0.05 ). Conclusion The expressions of GPIbα and vWF elevated in DVT patients and associated with the occurrence of VTE.
