摘 要:目的基于黄酮类化合物在碱性条件下可与钼酸钠形成稳定配合物的原理,比较研究标准芦丁和香椿叶总黄酮提取物与钼酸钠形成配合物的光谱特征,建立香椿叶总黄酮质量控制的新方法,考察不同产地香椿叶中总黄酮含量。方法香椿叶经甲醇连续提取得总黄酮提取液,其中的总黄酮与Na2MoO4-NaN02-NaOH反应形成稳定配合物。以芦丁作为标准,在200-500nm范围分别对芦丁配合物、香椿提取液总黄酮配合物与空白溶液进行差示扫描。结果标准芦丁配合物或香椿叶提取物总黄酮配合物与空白溶液在200-500nm范围的差示光谱图一致,且在332nm波长处呈现最大吸收峰。以芦丁为标准,在332nm波长测定,芦丁含量与吸光度在4~40μg·mL-1范围内符合比耳定律,其回归方程为A=0.0021+0.02804C,r=0.9994,方法的回收率为93.4%~100.1%。该法测定6个不同产地连续两年的香椿叶总黄酮的平均含量分别为(6.02±0.52)%和(6.50±0.32)%。结论钼酸钠配合物分光光度法测定香椿叶中总黄酮含量,操作简单易行,可较好解决背景吸收的问题,是一种实用的香椿叶总黄酮测定方法。Objective Based on the principle that the conjugates can be formed between flavonoids and Na2MoO4-NaNO2-NaOH, the spectroscopic characteristics of the conjugates of the total flavonoids in Toona sinensis leaves or authentic rutin with NazMoO4-NaNO2-NaOH were compared by a spectrometric method and the contents of the total flavonoids in Toona sinensis leaves were estimated. Methods Extractions of total flavonoids in the leaves of Toona sinensis were prepared by a consecutive extract with methanol. The stable conjugates were formed by adding the NazMoO4-NaNOz-NaOH solution in extractions of total flavonoids. With authentic rutin as standard, the conjugates of authentic rutin or those of total flavonoids with NazMoOn-NaNOz-NaOH were differential scanned using the wavelengths ranged from 200 nm to 500 nm by a UV-VIS spectrometer. Results The differential spectrogram of total flavoniods in Toona sinensis leaves with Na2MoO4-NaNO2-NaOH scanned in the range of 200 nm to 500 nm was similar as that of authentic rutin, with a maximum absorbance peak at 332 nm. The contents of total flavonoids in Toona sinensis leaves were determined with rutin as standard at 332 nm. Linear range of authentic rutin was 4-40μg.mL-1. The regression equation was A=0.002 1 + 0.028 04 C with a regression coefficient of 0.999 4. The rates of recovery of the method were 93.4%~100.1%. The average contents of total flavonoids in Toona sinensis leaves collected at six different spots in two successive years were (6.02~0.52)% and (6.50±0.32)%, respectively. Conclusions This is a simple and useful method for determining the total flavoniods in the leaves of Toona sinensis which can better solve the problem of background absorbance.
