摘  要:为鉴定广东水松的遗传多样性,应用改良CTAB法分别提取广东不同地区的孑遗植物水松基因组DNA,以真核生物rDNA ITS序列的通用引物分别对其ITS区进行PCR扩增及测序后进行比对、分析,将得到的序列申请登录入GenBank核苷酸数据库。结果表明:广东地区不同来源水松的rDNA ITS序列长度均为988bp,相互之间仅有2~3个碱基差异,说明该序列可作为从分子水平鉴别水松种质资源的参考标记。珠海、中山和广州地区孑遗植物水松GenBank核苷酸数据库登录号分别是KF977874、KF977876和KF977875。The genomic DNA of relic plant G.pensilis from different regions in Guangdong was extracted by the modified CTAB method,then the ITS of genomic DNA was amplified and sequenced by the universal primer of eucaryon rDNA ITS sequence respectively,and finally the obtained sequence by comparison and analysis was applied for entering GenBank.The results showed that the rDNA ITS sequence length of G.pensilis samples from different regions in Guandong is 988 bp and there is difference with 2-3 bases in sequence length between different G.pensilis samples,which means that the sequence can be used as the reference marker to identify Glyptostrobus pensilis germplasm resources based on molecular level. The accession number of G. pensilis from Zhuhai, Zhongshan and Guanzhou is KF977874,KF977876 and KF977875 in GenBank respectively.
