月舒滴丸对大鼠子宫内膜及平滑肌细胞OTR表达的影响

摘　　要：目的:探讨月舒滴丸对大鼠子宫平滑肌细胞缩宫素受体(Oxytocin receptor,OTR)基因表达及对大鼠子宫内膜上皮细胞前列腺素(Prostaglandin,PG)合成的影响,阐明月舒滴丸治疗原发性痛经的作用机制。方法:分别采用雌二醇(Estradiol,E2)和缩宫素(Oxytocin,OT)刺激,复制子宫平滑肌细胞OTR高表达模型,采用RTPCR法,探讨月舒滴丸对子宫平滑肌细胞OTR基因表达的影响;采用Elisa法观察月舒滴丸干预对大鼠子宫内膜上皮细胞PGF2α、PGE2含量变化的影响。结果:月舒滴丸能明显降低大鼠子宫平滑肌细胞OTR基因表达,并可以显著降低大鼠子宫内膜上皮细胞PGF2α水平,提高PGE2表达,降低子宫内膜细胞PGF2α/PGE2水平。结论:通过月舒滴丸的干预,可减轻缩宫素的刺激,从而使子宫平滑肌细胞OTR低表达,减少子宫平滑肌细胞对OT的敏感性,并且能够有效调节大鼠子宫内膜上皮细胞前列腺素水平。Objective：To explore the effects of Yueshu dropping pill on the gene expression of OTR on rats uterine smooth muscle cells and the content change of Prostaglandin（PG）expressed on rats endometrial epithelial cells（EC）,and to elucidate the mechanism of Yueshu dropping pill in treating primary dysmenorrhea.Methods：Uterine smooth muscle cells stimulated by E2 and OTR were used to establish high OTR expression model,and RT-PCR method was used to explore the effect of OTR expressed on uterine smooth muscle cells by Yueshuu dropping pill.ELISA was used to explore the content change of PGF2α and PGE2 expressed on rats endometrial epithelial cells.Results：Yueshu dropping pill reduces gene expression of OTR on rat uterine smooth muscle cells significantly.It reduces the content of PGF2α on endometrial epithelial cells and it enhances the content of PGE2.It also reduces the ratio of PGF2α/PGE2.Conclusion：Yueshu dropping pill reduces the stimulation of OT,and thus lowers the expression of OTR and reduces the susceptibility of uterine smooth muscle cells on OT.It also adjusts the level of PG on endometrial epithelial cells effectively.
