摘 要:目的:观察不同剂量冠心合剂对大鼠急性心肌梗死凋亡及线粒体途径相关凋亡蛋白表达的影响。方法:取雄性SD大鼠80只,随机分为模型组、冠心合剂高剂量组、中剂量组、低剂量组和对照组。连续喂养大鼠7 d后,结扎冠状动脉前降支,造成急性心梗模型。造模手术后4.5 h留取心脏标本,检测梗死面积、细胞凋亡指数以及Bax、Bcl-2、p-BAD、Caspase-8、Caspase-3蛋白表达量。结果:与模型组比较,冠心合剂组可明显减少心梗面积和心肌细胞凋亡,上调Bcl-2,p-BAD表达,减少Bax,Caspase-8和Caspase3蛋白表达。结论:冠心合剂抑制缺血导致的心肌梗死,减少心梗面积和心肌细胞凋亡,其机制与调节线粒体途径中相关凋亡蛋白的表达有关。Objective : To investigate the effects of Guanxin Heji on apoptosis in rats of acute myocardial infarction and expression of Bax, Bcl - 2, p - BAD, Caspase - 8 and Caspase - 3 in myocardium tissue. Methods : 80 male SD rats were randomly divided into 5 groups :model group, sham group, high- dose group, medium -dose group,low- dose group. After being administered corresponding medicine for 7 days, all groups except sham group underwent coronary ligation of left anterior descending branch to establish acute myocardial infarction models, and sham group only received thoracotomy without coronary ligation. Four hours after AMI, myocardium tissues were collected to detect the apoptotic index with TUNEL and protein expression of Bax, Bcl - 2, p - BAD, Caspase - 8 and Caspase - 3 by Western blot. Results : The infarction area and apoptotic index were reduced in Guanxin Heji group than that in model group, and protein expression of Bcl - 2 and P - BAD increased and Bax, Caspase - 8 and Caspase - 3 decreased significantly in Guanxin heji groups than those in model group. Conclusion:Guanxin Heji can reduce infarction area and inhibite the myocardial apoptosis by regulating Bcl - 2, Bax, P - BAD, Caspase - 8 and Caspase - 3 expressions.
