摘 要:目的:探讨西洋参茎叶总皂苷(PQS)对大鼠心肌缺血再灌注损伤后FOXO3a/Bim信号通路的影响。方法建立心肌缺血再灌注模型(IR组),心肌缺血再灌注+PQS预处理组(IR+P)在术前3 d腹腔注射PQS,假手术组(S组)仅接受假手术。再灌注24 h后TUNEL法检测心肌细胞凋亡、生化指标测定、免疫组化和Western blots检测心肌FOXO3a,p-FOXO3a和Bim蛋白表达,实时荧光定量PCR检测心肌Bim mRNA表达。结果:TUNEL结果显示,与S组(0.6±0.0)相比,IR组(23.1±3.7)和(IR+P)组(12.4±3.1)凋亡率明显升高(P<0.01),且(IR+P)组明显低于IR组(P<0.01)。IR组和(IR+P)组心肌SOD明显低于S组(P<0.01),且(IR+P)组明显高于IR组(P<0.01);IR组和(IR+P)组心肌MDA、CK、LDH高于S组(P<0.05或P<0.01),且(IR+P)组明显低于IR组(P<0.05或P<0.01)。免疫组化和Western blots结果发现,三组心肌组织FOXO3a蛋白表达差异没有统计学意义(P>0.05);IR组和(IR+P)组p-FOXO3a蛋白明显低于S组(P<0.01),(IR+P)组高于IR组(P<0.01);IR组和(IR+P)组Bim蛋白明显高于S组(P<0.05或P<0.01),(IR+P)组低于IR组(P<0.05或P<0.01)。实时荧光定量PCR结果显示,与S组比较,IR组和(IR+P)组Bim mRNA明显升高(P<0.01),(IR+P)组低于IR组(P<0.01)。结论西洋参茎叶总皂苷可能是通过基因转录和蛋白质翻译水平调节FOXO3a/Bim信号通路对心肌缺血再灌注损伤起到保护作用。Objective: To investigate the effects of Panax quinquefolium saponins on FOXO3a/Bim signaling pathway in myocardial ischemia- reperfusion injury (MIRI) rats. Method: The myocardial I/R injury model in rats was established (IR group). The myocardial I/R + PQS preconditioning group (IR + P) was intraperitoneally injected PQS for preoperative 3 d and the sham operation group (S group) received only sham surgery. Cardiomyocyte apoptosis was detected by TUNEL assay and myocardial biochemical parameters were measured. The expressions of FOXO3a, p - FOXO3a and Bim protein in myocardial were detected by immunohistochemistry and Western blots. The expressions of myocardial Bim mRNA were detected by real - time PCR after reperfusion 24 h. Results : TUNEL results showed that compared with the S group ( 0.6 ± 0.0 ), IR group ( 23.1 ± 3.7 ) and ( IR + P) group ( 12.4 ± 3.1 ) were significantly increased apoptosis ( P 〈 0. 01 ) and ( IR + P) group' s was significantly lower than that of the IR group ( P 〈 0.01 ). Immunohistochemistry and Western blots results showed that the difference of FOXO3a protein expressionof three groups myocardial was not statistically significant ( P 〉 0.05).
