摘 要:目的:研究蝉翼藤有机酸类提取物(CYTYJS)对鸭乙型肝炎病毒(DHBV)复制的抑制作用与保肝的作用。方法:将DHBV DNA阳性的广西麻鸭,随机分成5组,分别为CYTYJS高、中、低剂量(20,10,5mg·kg-1·d-1)组,模型组和拉米夫定(3TC)组。于用药前(T0)、用药7 d(T7)、14 d(T14)、21 d(T21)及停药后3 d(P3)分别采血,检测血清中DHBV DNA的滴度、丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST)的含量,实验结束时,取肝组织做病理学检查。结果:与模型组比较,CYTYJS各剂量组用药后血清DHBV DNA滴度显著降低(P<0.05或0.01),停药3 d后,CYTYJS各剂量组DHBV DNA,ALT和AST仍在较低水平(P<0.05)而阳性组则有明显回升现象;镜下观察各组肝脏组织切片,与模型组比较CYTYJS高、中、低剂量组肝脏的病理改变有明显改善。结论:CYTYJS有一定的抗乙肝病毒和保肝的作用。Objective: To determine the inhibiting effect of organic acids extracted from the securidaca in- appendiculata (CYTYJS) on DHBV replication and the protective effect on liver function. Methods: The DHBV- DNA positive ducks from Guangxi Zhuang Autonomous Region were randomly divided into five groups, and treated with CYTYJS (20, 10 and 5 mg kg-1 d-1) or lamivudine (positive control). Before treatment (T0), seventh days ( T7 ) , fourteenth days (T14) and twenty-first days ( T21 ) after treatment, and third days after drug withdrawal (P3) , blood samples were collected to determine the serum contents of DHBV-DNA, ALT and AST, respectively. At the end of the experiment, histopathological changes of the liver were examined. Results: After treatment with CYTYJS, the serum DHBV DNA titer significantly decreased compared with the model group (P 〈 0.05 or 0.01 ). At the 3th day after withdrawal, the titer of DHBV DNA, and the levels of ALT and AST in each CYTYJS group were still in a lower level, while there was an obvious rebound phenomenon in the positive control (lamivudine). The light microscopic examination of liver biopsy showed that pathological changes were significantly improved in CYTYJS groups and the positive control group compared with model group. Conclusion: CYTYJS have the anti- hepatitis B virus and hepatoprotective effect.
