丹参粉针剂对酒精致大鼠慢性肝纤维化的保护作用

摘　　要：目的:研究丹参粉针剂对酒精性肝纤维化模型大鼠的保护作用。方法:Wistar大鼠随机分为正常组、模型组、阳性对照组(水飞蓟素混悬液50 mg·kg-1)和丹参粉针剂低、中、高剂量组(25,50,100 mg·kg-1)。除正常组外,各组每天给予白酒-玉米油-吡唑混合液灌胃以建立酒精性纤维化模型,同时分别给予相应治疗药物。9周后眼眶静脉丛取血,分离血清,测定丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性及白蛋白(ALB)、胆固醇(CHOL)、低密度脂蛋白胆固醇(LDL-C)、透明质酸(HA)、III型前胶原(PCIII)含量;处死大鼠,灌流,取肝脏,进行肝组织病理HE、天狼猩红染色。结果:丹参各剂量组大鼠肝脏体积明显小于模型组,质地颜色有所恢复;丹参各剂量组肝功能指标ALT,AST活性、血脂指标CHOL,LDL-C及纤维化指标HA,PCIII含量显著降低(P<0.05或0.001),ALB含量显著回升(P<0.001)。肝组织病理HE染色显示丹参各剂量组脂肪变性显著减少,细胞形态有所恢复,坏死及炎细胞少见。天狼猩红染色显示胶原纤维显著减少。结论:丹参粉针剂对酒精性肝纤维化模型大鼠具有保护作用。Objective： To determine the protective effect of Danshen powder for injection on rats with alco- holic liver fibrosis. Methods： Wistar rats were randomly divided into normal, model, positive control and three doses of Danshen powder for injection groups. Except for those in nomal groups, the rats were administered a mixture of alcohol （8 - 12 g.kg-1）, corn oil （2 gkg-1 ） and pyrazole （24 mgkg-1 ） by garage every day for 9 weeks to induce alcoholic liver fibrosis. Meanwhile, the rats in the drug treatment groups were given silymarin （50 mg kg - 1 ） by garage or Danshen powder for injection （25, 50, 100 mg.kg-1） by intraperitoneal injection. At the end of the experiment, blood was taken from each rat under orbit, and serum was separated. The protective effect was evaluated by determination of serum aspartate transaminase （AST） , alanine transaminase （AST） , albumin （ALB） , cholesterol （CHOL） , low density lipoprotein cholesterol （ LDL-C）, hyaluronic acid （HA） , and type III procollagen （ PCIII）, as well as observation of liver pathology by hematoxylin eosin （HE） and sirius red staning. Results ： Danshen powder for injection treatment ameliorated the liver characteristics. Serum ALT, AST, HA, and PCIII were reduced while ALB was improved. The pathology examination showed that Danshen powder for injection reduced hepatic steatosis, necrosis, and inflammation and improved the state of hepatocytes. Conclusion： Danshen powder for injection can protect rats from alcoholic liver fibrosis.
