摘 要:目的:考察热毒宁注射液(RDN)对脂多糖(LPS)致大鼠急性肺损伤(ALI)的治疗作用及其机制研究。方法:SD大鼠60只,将大鼠随机分成6组,每组10只,即正常组、模型组、阳性药组(地塞米松2 mg·kg-1)、热毒宁注射液高、中、低剂量(10.16,5.08,2.54 g·kg-1)组。除正常组外采用LPS按6 mg·kg-1对SD大鼠气管内滴注制作急性肺损伤模型,观察受试药热毒宁注射液3个剂量组和阳性药地塞米松注射液(DEX)iv给药后对肺脏湿/干重(W/D)、支气管肺泡灌洗液(BALF)总蛋白含量和血清超氧化物歧化酶(SOD)、髓过氧化物酶(MPO)活性,丙二醛(MDA)含量及肺组织匀浆白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)含量的影响。结果:与正常组比较,模型组大鼠W/D升高、BALF总蛋白含量升高、MDA含量和MPO活性升高,SOD活性降低,IL-1β,TNF-α,IL-10水平升高(P<0.01),与模型组比较,热毒宁注射液不同剂量均降低大鼠W/D,降低BALF总蛋白含量、降低MDA含量和MPO活性,增强SOD活性,降低IL-1β,TNF-α,IL-10水平(P<0.01,P<0.05)。结论:RDN可改善肺组织病理变化,减轻LPS致ALI过程中体内的氧化应激反应,减少炎性因子可能是治疗急性肺损伤疾病的重要机制。Objective: To investigate the mechanism and the rapeutic effect of Reduning injection( RDN) on acute lung injury( ALI) in rats. Method: Sixty rats were randomly divided into six groups,normal group,model group,positive control group( dexamethasone,2 mg·kg^-1),high-,medium-,low-dose RDN groups( 10. 16,5. 08,2. 54 g·kg^-1). The ALI rats was induced by ipopolysaccharide( LPS) except for rats in normal group. The effects of wet / dry weighto( W / D) ratio in lung,the total protein contents of bronchoalveolar lavage fluid( BALF),superoxide dismutase( SOD),malondialdehyde( MDA),myeloperoxidase( MPO) in serum and interleukin( IL)-1β,tumor necrosis factor( TNF)-α,IL-10 in lung homogenate were observed. Result: The W /D ratio,BALF,the content of MDA,IL-1β,TNF-α,IL-10 were decreased,the activity of MPO was inhibited and the activity of SOD was activated in model group. The above indexes were all improved in all dosage of RDN groups( P〈0.01,P〈0.05). Conclusion: RDN can improve the pathological changes of lung tissueand reduce the oxidative stress reaction of ALI process. Its mechanism may be achieved by reducing the inflammatory factor in vivo.
