摘 要:目的:探讨茵陈蒿汤对阻塞性黄疸大鼠i NOS表达异常引起肝损伤的保护作用机制,为临床围手术期合理应用中药提供理论依据。方法:选择成年SD大鼠48只,随机分组为1阻塞性黄疸再通模型组(OJR组)24只;2阻塞性黄疸再通模型+中药组(OJRC组)24只。分别于胆道梗阻7 d后解除梗阻,并于解除梗阻后1,3,7 d每个时相点处死大鼠8只。检测血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、血清总胆红素(TBIL);观察肝组织i NOS mRNA及蛋白的表达情况。结果:茵陈蒿汤能够明显降低大鼠血清ALT,TBIL含量(P<0.05),抑制肝组织i NOS mRNA及i NOS蛋白的表达(P<0.05)。结论:茵陈蒿汤可抑制肝脏i NOS的表达,从而促进阻塞性黄疸大鼠梗阻解除后肝功能的恢复。Objective: To explore the protective mechanism of Yinchenhao decoction in liver injury which caused by abnormal expression of i NOS in obstructive jaundice rats. And to provide the theory basis for reasonable use traditional Chinese medicine during perioperative period. Method: Select 48 adult SD rats,randomly divided into two groups1 24 rats in obstructive jaundice reperfusion model group( group OJR),2 24 rats in obstructive jaundice reperfusion model plus traditional Chinese medicine group( OJRC group). Relieve obstruction of the two groups in day 7 after the biliary tract obstructed and put rats to death in day 1,3,7 after the obstruction removed.Detect serum alanine aminotransferase( ALT),aspartate aminotransferase( AST),serum total bilirubin( TBIL),observe the expression of i NOS mRNA and i NOS protein in the liver tissue. Result: Yinchenhao decoction can significantly reduce the content of serum TBIL,ALT( P〈0.05),and inhibit i NOS expression of mRNA and i NOS protein in rats liver tissue( P〈0.05). Conclusion: Yinchenhao decoction can inhibit the expression of hepatic i NOS,so as to promote the recovery of liver function in rats with obstructive jaundice.
