摘 要:目的 :研究中药复方芎汤提取物及含药血清对人肝癌细胞BEL-7402活力的影响。方法 :分别给SD大鼠灌服芎汤超临界萃取部位(SFE)和大孔树脂分离得到的20%、40%乙醇洗脱部位(EE20、EE40部位),获得各部位含药血清,设置含药血清组、提取物组、顺铂对照组、空白对照组分别作用于人肝癌细胞BEL-7402,用MTT法测定各组对人肝癌细胞BEL-7402活力的影响。结果:芎汤EE20含药血清在低浓度时抑制作用明显,并随着浓度增高抑制作用减弱;EE40部位含药血清随着药物浓度的增大对癌细胞增殖作用也越来越强,SFE部位含药血清对细胞无显著性作用;提取物对细胞的作用趋势与含药血清基本一致,但含药血清作用强于提取物。结论:芎汤EE20有效部位在低浓度时对人肝癌细胞BEL-7402的增值有一定的抑制作用,为其抗肿瘤作用的进一步研究提供了依据。Objective:To research the effect of active part of Xiongqiong decoction and its medicine-contained serum on the vitality of BEL-7402 human hepatocellular carcinoma cells. Methods:Fed SD rats with SFE part,EE20 part and EE40 part separated by Macroporous resin of Xiongqiong decoction respectively to get medicine-contained serum of every part. The BEL-7402 human hepatocellular carcinoma cells were acted upon by medicine-contained serum,extract,Cisplatin and RPMI-1640. The effect on vitality of BEL-7402 human hepatocellular carcinoma cells was detected by MTY method. Results:The inhibitory effect of serum containing EE20 of Xiongqiong decoction was significant at low concentration and it weakened as the concentration increased. The effect of EE40-contained serum on the proliferation of cancer cells strengthened as the drug concentration increased. The SFE-contained serum had no significant effect on cells. The effect of extract on cells was basically the same as that of the medicine-contained serum,but it was weaker than that of the medicine-contained serum. Conclusion:The active part EE20 of Xiongqiong decoction had some inhibitory effect on the proliferation of BEL-7402 human hepatocellular carcinoma cells at low concentration. This provides a basis for the study of its anti-tumor effect.
