摘 要:目的观察针刺捻转补泻对应激性高血压大鼠下丘脑及血浆中内皮素(ET)、血清去甲肾上腺素(NE)、5-羟色胺(5-HT)的影响,探讨捻转补泻手法对应激性高血压大鼠血压调控的作用机制,为捻转补泻效应差异提供客观依据。方法将60只正常雄性Wistar大鼠随机等分为5组,每组12只,分别为:正常组,模型组,针刺对照组,针刺补法组,针刺泻法组。除正常组外,各组采用电击足底结合噪声刺激法制备应激性高血压大鼠模型。除正常组、模型组外,其余各组分别用不同的手法针刺"太冲"穴1周后,取血、取脑,采用ELISA法测定血清中NE、5-HT含量,采用放射免疫法测定血浆及下丘脑中ET含量,比较各组间指标有无差异。结果与正常组比较,模型组血清NE、5-HT,血浆及下丘脑ET含量明显升高,有显著差异(P<0.05);针刺对照组与模型组比较无统计学差异(P>0.05)。针刺泻法组血清NE、5-HT、血浆及下丘脑ET含量较模型组、针刺对照组、针刺补法组明显降低,有显著差异(P<0.05),和正常组比较无统计学差异(P>0.05)。结论针刺泻法可使应激性高血压大鼠血清NE、5-HT含量下降,血浆及下丘脑ET含量下降。针刺对应激性高血压大鼠血压调节与中枢和外周ET、血清NE、5-HT含量变化有关,其机制是外周交感神经系统、血管内皮功能系统共同作用的结果。Objective To explore the potential mechanism of twirling reinforcement-reduction method of acupuncture in rats with stress-induced hypertension by investigating the effects on plasma concentration of endothelin ( ET), norepinephrine ( NE ) and 5-hydroxytryptamine (5-HT). Methods Overall 60 male Wistar rats were randomly divided into normal group, model group, acupuncture control group, twirling reinforcement acupuncture group and twirling reduction acupuncture group, 12 rats in each group. The model of stress-induced hypertension was established by foot-shock and noise stimulation. All three acupuncture groups were treated with acupuncture in left Taichong (LR3) with corresponding methods. The plasma concentrations of ET, NE and 5-HT and hypothalamic concentration of ET were detected after 7 d. Results Compared with normal group, concentrations of ET, NE and 5-HT in blood plasma and hypothalamus in model group increased significantly ( P 〈 0.05 ). There is no statistical differences between acupuncture control group and model group ( P 〉 0.05 ). Compared with model group, concentrations of ET, NE and 5-HT decreased significantly in twirling reduction acupuncture group(P 〈 0.05 ), even, decreased to the levels of normal group (P 〉 0.05 ) ; however, those tested showed no statistical differences in acupuncture control group and twirling reinforcement group. Conclusion Twirling reduction method of acupuncture decreased the concentration of ET, NE and 5-HT in rats with stress- induced hypertension. The regulation of blood pressure is associated with concentrations of of central and peripheral ET, NE and 5-HT, which is maybe the interaction between peripheral sympathetic nervous system and vascular endothelial system.
