摘 要:目的利用响应面分析法对秀丽莓总糖提取工艺进行优化。方法以秀丽莓水提液总糖含量为评价指标,选取提取温度(X1)、提取时间(X2)、料液比(X3)、药材粉碎粒径(X4)及提取次数(X5)为单因素进行考察,最终确定提取温度、提取时间及料液比为主要影响因素。在该试验基础上选取3个因素的3个水平,根据中心组合(Box-Benhnken)试验设计原理,采用响应面分析法,以水提液的总糖含量为响应值(Yyielded)作响应面及等高线,计算获得可反应3因素与总糖含量之间关系的多元二次线性回归方程,优化设计最佳提取工艺。结果根据计算拟合所得方程确定的最佳提取工艺条件为:药材粉碎过40目筛,浸提温度65℃,提取时间2.5 h,按料液比1∶21加水提取1次。按照该条件提取所得秀丽莓水提液总糖含量可达13.39%,与理论预测值13.77%基本一致。结论该最佳提取条件可靠、稳定、提取率高,基本能满足工业生产的需求。Objective To approach an optimal extraction procedure of saccharides from Rubus amabilis by using response surface methodoloy. Methods With the saccharides content of water extract as an evaluation index, single-factor experiment was conducted on 5 factors respectively, including extraction temperature ( X1 ) , extraction time ( X2 ) , solvent ratio ( X3 ) , crush size ( X4 ) and extraction times (X5). 3 factors (X1, X2, X3 ) were determined to be crucial. According to the central composite experimental design principles(Box-Benhnken) , three different levels of each of 3 factors selected was adopted by response surface methodology (saceharides content was chosen as response value, while response surface and contour curves were plotted ), in search for multiple quadratic linear regressions between the crucial 3 factors and saccharides content. Results The optimum extraction conditions of the saccharides from Rubus amabilis were calculated from the formula above as the following: powdered herbs filtrated through 40 meshes, extraction temperature was set at 65 ℃, extraction time was 2.5 hours with 21 times of water (w/v) and extracted once. Under these conditions, the content of saccharides from Rubus amabilis was up to 13.39%, which was in accordance with the theoretical prediction 13.77%. Conclusion The extraction condition optimized was reliable, stable and effective, which can meet with the requirement of industrial production.
