摘  要:目的探讨大鼠吸入氯气急性肺损伤(ALI)肺组织基质金属蛋白酶-9(MMP-9)表达的变化。方法将40只雄性SD大鼠随机分为对照组和4个不同浓度[(10.28±1.73)、(42.00±3.18)、(102.00±6.96)、(160.14±6.87)mg/m3]氯气染毒组,每组8只,动式吸入染毒10min,6h后行血气分析,取肺组织测湿干重比(W/D)和病理切片观察,用ELISA方法测支气管肺泡灌洗液(BALF)中MMP-9蛋白表达水平,用RT-PCR方法测肺组织MMP-9mRNA相对表达。结果与对照组比较,各染毒组大鼠BALF中MMP-9蛋白和肺组织MMP-9mRNA水平均有升高,差异有统计学意义(P<0.05);且随着氯气染毒浓度的增加,大鼠BALF中MMP-9蛋白和肺组织MMP-9mRNA水平均持续升高,差异有统计学意义(P<0.05)。结论 MMP-9表达改变可能是大鼠吸入氯气致ALI重要的发病机制之一。Objective To explore the changes of gene and protein matrix metalloproteinase-9(MMP-9)in rats exposed to different concentrations of chlorine.Methods Forty male SD rats were randomly divided into one control group and four experiment groups inhaled different concentration of chlorine for 10 minutes each,and were sacrificed at time point of 6hs after cessation of exposure.The concentration of MMP-9in the bronchoalveolar lavage fluid(BALF)by ELISA,and the expression of MMP-9mRNA in lung tissue by reverse transcription-polymerase chain reaction(RT-PCR)were analyzed,while the detection of the content of PaO2,wet/dry weight ratio of lung tissue(W/D)and the pathologic observation were taken respectively.Results The expression of MMP-9in BALF and lung tissue was increased significantly in all experiment groups as compared with the control group(P〈0.05),and gradually increased with the concentration of chlorine rises(P〈0.05).Conclusions MMP-9may play an important role in progression of acute lung injury induced by chlorine.
