球松素对脂多糖诱导的人脐静脉内皮细胞损伤的保护作用

摘  要:目的观察球松素对人脐静脉内皮细胞的保护作用。方法采用灌注消化法分离培养人脐静脉内皮细胞(HUVEC),并对其进行形态学和免疫组织化学染色鉴定。用脂多糖(LPS)诱导建立人脐静脉血管内皮细胞的损伤模型,随后用球松素作用于受损伤细胞,观察其对HUVEC的保护作用。结果倒置显微镜下可见细胞呈梭形,单层铺路石状排列,Factor VIII相关抗原免疫组织化学染色鉴定阳性。100μg/L的LPS对脐静脉内皮细胞生长有明显的抑制作用,可作为人脐静脉内皮损伤模型的造模浓度。球松素能减轻LPS对细胞的损伤(P<0.05),并呈剂量依赖关系。结论球松素对LPS所诱导的人脐静脉内皮细胞损伤有保护作用。Objective:To observe the protective effect of Pinostrobin on human umbilical vascular endothelial cell(HU-VEC). Methods:HUVEC were collected by digestion - perfusion culture and identified by morphology and Immunohisto-chemistry?method. Then LPS was used to induce the damage and the injury model was established. Finally,the protective effect of Pinostrobin on HUVEC was observed. Results:Under the inverted microscope,cells were spindle - shaped paving stones arranged in single layer. It was positive for Immunohistochemical staining of Factor VIII - related antigen. LPS (100μg/ L)inhibited the growth of HUVEC,and could be used as the concentration of the model. Pinostrobin reduced the damage of LPS in a dose - dependent manner(P 〈 0. 05). Conclusion:Pinostrobin can protect the HUVEC injury by LPS.
