摘 要:目的:建立臭蒿中东莨菪内酯成分的HPLC定性定量分析方法。方法:色谱柱为Agilent TC-C18柱(4.6 mm×250 mm,5μm),柱温30℃,流动相为甲醇(A)-0.3%磷酸溶液(B)梯度洗脱,流速1.0 m L·min-1,检测波长344 nm。结果:东莨菪内酯质量浓度与峰面积在测定范围内呈良好的线性关系(r=0.999 9),重现性好,RSD值为1.51%;平均加样回收率为101.42%。结论:本试验建立的HPLC方法稳定可靠、简便易行,为臭蒿药材的质量评价和质量控制提供了理论依据。In this paper, a qualitative and quantitative analysis method was established for the scopoletin in herba Artemisiae hedinii hy HPLC. The chromatographic column was Agilent TC-C18(4.6 mm×250 mm, 5 μm); the temperature of column was 30℃, eluted with a mobile phase consisted of methanol and 0.3% phosphoric acid solution and gradient elution at a flow rate of 1.0 m L·min^-1. The detection wavelength was 344 nm. The results showed that the mass concentration and peak area of scopoletin had a good linear relationship within the range of measurement(r=0.999 9). The reproducibility was good; and the RSD was 1.51%. The average sample recovery rate was 101.42%. It was concluded that the established method was stable, reliable and simple. It provided theoretical evidences for the quality evaluation and quality control of herba A. hedinii.
