摘 要:目的:研究2,4-二硝基氯苯( DNCB)诱导的超敏反应性皮炎小鼠淋巴细胞增殖与活化情况。方法无特定病原体级雌性C57BL/6小鼠随机分为对照组、低剂量组和高剂量组,每组6只。第1~3天3组小鼠分别采用100μl质量体积比为0.00%、0.50%和1.00%的DNCB进行背部涂抹诱导,第4天不作处理,第5天分别采用25μl质量体积比为0.25%、0.25%和0.50%的DNCB进行左耳耳廓激发,24 h后观察耳廓过敏症状。流式细胞术检测脾脏和耳后淋巴结淋巴细胞增殖、活化和亚群。结果高剂量组小鼠左耳耳廓肿胀,血管扩张;对照组和低剂量组均未观察到上述症状。高剂量组耳廓肿胀度大于对照组[(5.30±2.04)vs(0.50±0.49)mg,P〈0.05]。低、高剂量组脾脏系数和耳后淋巴结脏器系数均高于对照组[脾脏:(0.560±0.040)% vs(0.420±0.120)%,(0.590±0.070)% vs(0.420±0.120)%,P 〈0.05;耳后淋巴结:(0.018±0.002)% vs(0.005±0.005)%,(0.021±0.007)% vs(0.005±0.005)%,P〈0.05]。低、高剂量组小鼠脾脏和耳后淋巴结中T淋巴细胞增殖比例均高于对照组[脾脏:(1.45±0.35)% vs(0.65±0.35)%,(1.77±0.12)% vs(0.65±0.35)%,P 〈0.05;耳后淋巴结:(1.40±0.36)% vs(0.50±0.10)%,(1.30±0.26)% vs(0.50±0.10)%,P〈0.05]。高剂量组脾脏CD44+细胞比例高于对照组[(95.25±1.25)% vs(92.57±1.19)%,P 〈0.05],CD62L+细胞比例低于对照组[(22.93±2.10)% vs(44.80±2.36)%,P 〈0.05]。低剂量组小鼠耳后淋巴结 T 淋巴细胞比例、CD4+细胞比例和CD4+/CD8+比值均低于对照组[(29.70±1.83)% vs(39.40±2.53)%,(20.30±2.60)% vs(31.30±2.84)%,(2.64±0.03)vs(5.29±1.17),P〈0.05]。高剂量组小鼠脾脏T淋巴细胞比例、CD4+细胞比例和CD8+细胞比例均低于对照组[(15.55±0.51)% vs(25.70±1.47Objective To investigate the lymphocyte proliferation and activation in the 2,4-dinitrochlorobenzene( DNCB)-induced hypersensitivity dermatitis mice model. Methods Specific pathogens free 8-12 week-old female C57BL/6 mice were randomly divided into control group,low- and high-dose groups,6 mice per group. Mice in 3 groups were induced daily with 100 μl mass-to-volume ratio of 0. 00%,0. 50% and 1. 00% DNCB respectively on back skin on day 1-3,no special treatment on day 4,and then stimulated once on the dorsum of left ears on day 5 with 25μl mass-to-volume ratio of 0. 25%,0. 25% and 0. 50% DNCB respectively,24 hours later the auricle allergy symptoms were observed. Flow cytometry was adopted to detect the lymphocyte proliferation,activation and subpopulations of spleens( SPs)and auricular lymph nodes( ALNs). Results The left ears of mice were swelling and angiectatic in high dose group,while symptoms were not observed in the control and low dose groups. The grade of ear swelling in high dose group was higher than that in the control[(5. 30 ± 2. 04)vs(0. 50 ± 0. 49)mg,P〈0. 05]. Both T lymphocyte proliferation proportions of SPs and ALNs in low- and high-dose groups were significantly higher than those of the control[SPs:(1. 45 ± 0. 35)% vs(0. 65 ± 0. 35)%,(1. 77 ± 0. 12)% vs(0. 65 ± 0. 35)%,P〈0. 05;ALNs:(1. 40 ± 0. 36)% vs(0. 50 ± 0. 10)%,(1. 30 ± 0. 26)% vs(0. 50 ± 0. 10)%,P〈0. 05].
