摘 要:本试验选用MHCⅡ、CD11b和CD16三种猪树突状细胞(DC)的抗体,对10头2月龄的杜长大三元杂交猪咽鼓管扁桃体和软腭扁桃体中的DC进行标记,应用荧光显微镜和激光共聚焦显微镜技术对分布于猪扁桃体内的DC进行观察和分析。结果发现:猪咽鼓管扁桃体和软腭扁桃体黏膜下固有层和淋巴滤泡内的DC可共表达CD11b与CD16或CD11b与MHCⅡ分子。咽鼓管扁桃体和软腭扁桃体黏膜下固有层中双阳性DC数量大约是淋巴滤泡双阳性DC数量的3倍。正常状态下双阳性DC在扁桃体上皮细胞间分布较少,咽鼓管扁桃体上皮细胞间几乎没有分布。单阳性CD11b+DC多分布于咽鼓管上皮细胞下方,仅有少量的CD11b+DC可向上皮细胞方向伸出胞质突起。软腭扁桃体和咽鼓管扁桃体固有层内分布有一定数量的DC,有利于摄取抗原和诱导免疫应答,为猪鼻腔免疫和口服免疫提供理论依据。The purpose of this article was to investigate the distribution of dendritic cells( DC) in healthy porcine tonsil. In this study,three different anti-porcine DC antibodies and fluorescence microscopy as well as confocal microscopy were used to examine the distribution of porcine tonsil DC on 10 crossed-bred( Duroc×Landrace×Yorkshine) pigs aged 2 months. The results showed that: DC in porcine tubal tonsil and soft palatine tonsil could co-express CD11 b with CD16 or CD11 b with MHCⅡ; The number of doublestained DC in porcine tonsil lamina propria were three times higher than lymphoid follicles; There were only small amounts of DC between the epitheliums,and DC in soft palatine tonsil were hardly be found; The majority of CD11b^+DC were observed under the epithelium of tonsil; A rare population of entending cytoplasmic processes between epithelium had been identified as a possible correlate for antigen uptake. There were cetanin number of DC in lamina propria of porcine tubal tonsil and soft palatine tonsil for antigen absorption and immune activation which provided a theoretical basis for intranasal and oral immunization.
