不同鸭种肌肉发育早期TRα基因的表达及其与部分生长性状的相关分析

摘  要:为探讨不同鸭种胚胎期和初生早期胸肌和腿肌中甲状腺激素受体(TRα)基因表达水平的发育性变化及其对胸肌质量、腿肌质量和胚质量(或体质量)的影响,选用生长速度差异较大的高邮鸭和金定鸭为研究材料,采用实时荧光定量PCR方法检测两鸭种13、17、21、25、27胚龄和出雏后7日龄时胸肌和腿肌中TRαmRNA的表达水平。结果表明:在鸭胚胎期和初生早期,两鸭种肌肉TRαmRNA的表达趋势较为一致,即在胸肌中呈"W"形,在腿肌中呈前高后低的表达规律。在13、17和21胚龄时两鸭种胸肌TRαmRNA的表达量均极显著低于腿肌(P<0.01),而在25、27胚龄和7日龄时则均高于腿肌。相关性分析表明:两鸭种胸肌中TRαmRNA的表达量与其胸肌质量、胚质量(或体质量)均呈极显著正相关(P<0.01),腿肌中TRαmRNA的表达量与其腿肌质量、胚质量(或体质量)均呈极显著负相关(P<0.01)。提示:肌肉中TRα基因表达具有显著的年龄依赖性和组织特异性,但不存在品种差异;TRα可能对鸭肌肉的生长发育具有一定的作用。The present study was designed to detect the developmental changes of thyroid hormone receptor alpha A( TRα) gene expression in pectorales muscle and legs muscle in two duck breeds with differential growth rate during embryonic and neonatal period and their effects on weight of pectorales,legs and body. Real-time PCR was applied to detect the expression level of TRαmRNA in pectorales muscle and legs muscle of Gaoyou ducks and Jinding ducks at the embryonic day 13,17,21,25,27 and 7 dayold. The results showed that TRα mRNA expression profiles have a similar trend in two breeds in the same muscle tissue which show " W" trend in pectorales muscle and high to low trend in legs muscle. The expression levels of TRα mRNA in pectorals muscle of two duck breeds are both significantly lower than those in legs muscle at 13,17,21 embryo age( P〈0. 01). While the expression levels of TRα mRNA in pectorals muscle in two duck breeds are both higher than those in legs muscle at 25,27 embryo age and 7 dayold. The correlation analysis shows that in both two duck breeds the expression levels of TRα mRNA in pectorals muscle are highly significantly positively correlated with weight of pectorals muscle and body( P〈0. 01),while the expression levels of TRα mRNA in legs muscle are highly significantly negatively correlated with weight of legs muscle and body( P〈0. 01). These results indicate that the expression of TRα gene is significantly affected by age and organization,but not affected by breed and TRα may play an important role in the muscle growth and development in duck.
