摘  要:目的 观察中草药半边旗提取物5F对三阴型乳腺癌细胞株增殖、凋亡的影响及探讨其机制.方法 不同浓度的5F(0、5、10、20、40 mg/L)分别处理MDA-MB-231乳腺癌细胞,采用细胞计数试剂盒(CCK-8)法检测5F对乳腺癌细胞株增殖的影响;荧光显微镜下观察5F处理后细胞形态的变化;流式细胞仪检测5F处理后乳腺癌细胞株的凋亡率;收集5F处理后的乳腺癌细胞株,采用Western blot方法检测细胞凋亡相关蛋白[B细胞淋巴瘤/白血病-2(bcl-2)、bel-2相关X蛋白(bax)、半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-3]表达水平.结果 5F可抑制MDA-MB-231乳腺癌细胞的增殖,其抑制效果与5F的浓度及作用时间呈正相关(P<0.05);流式细胞仪检测显示,经5F作用24 h后,MDA-MB-231细胞的凋亡率为40.13%,较阴性对照组明显增加(P<0.05);Western blot检测发现,经5F处理后,参与细胞凋亡线粒体途径的抗凋亡蛋白bcl-2表达减弱,促细胞凋亡蛋白bax表达增强,凋亡执行分子Caspase-3表达增强,且上述蛋白表达水平与5F呈明显的剂量依赖性(P<0.05).结论 5F可抑制三阴型乳腺癌细胞的增殖,抑制效果与5F浓度和作用时间相关;其作用机制之一可能是通过诱导细胞凋亡实现的.Objective To explore the anti-cancer effects and its mechanisms of ent-11 α-hydroxy-15-oxo-kaur-16-en-19-oic-acid (5F),a compound isolated from pteris semipinnata L (PsL),on human triple-negative breast cancer cells.Methods Triple-negative human breast cancer cells were treated with different concentrations of 5F (0,5,10,20,40 mg/L) for different periods respectively.Cell viability was detected by a cell counting kit-8 (CCK-8).Morphologic changes were examined under a fluorescence microscope after treatment with 5F for 24 h.The appotosis rate of cells was examined by flow cytomety.The expression of representative apoptosis-related proteins was detected by Western blotting.Results 5F inhibited the proliferation of triple-negative breast cancer cells in a concentration-and time-dependent manner (P 〈 0.05).The appotosis rate of malondialdehyde (MDA)-MB-231 was 40.13%,significantly higher than the control group (P 〈 0.05).Western blotting revealed that after treatment with 5 F,the expression of anti-appototic protein B cell lymphoma/leukemia-2 (bcl-2) in breast cancer cells was decreased,and that of pro-appototic protein bcl-2 associated X protein (bax) and Caspase-3 was increased.Conclusion 5F could inhibit the proliferation of triple-negative breast cancer cells in a concentration-and timedependent manner probably by inducing cell apoptosis.
