摘 要:目的研究药物浓度、灌流速度及实验方法对同心圆和线性2种微透析探针回收率的影响。方法以苦参碱为实验药物,分别测定药物不同浓度、不同灌流速度下,2种探针体外正向、反向回收率及体内反向回收率。结果探针回收率均与药物浓度无关,与灌流速度成反比;2种探针体外正向、反向回收率具有一致性,且均大于体内反向回收率;体内回收率同心圆探针大于线性探针。结论对于苦参碱的药动学研究,体外正向透析法简便准确,测得的回收率可作为药物的探针回收率。OBJECTIVE To evaluate the effects of drug concentration, perfusion rate and microdialysis method on the recoveries of concentric cannula and linear microdialysis probes. METHODS Matrine was selected as the model drug. The in vitro recovery was determined using positive dialysis and retrodialysis methods at different perfusion rates and concentrations. And the in vivo recovery was determined using retrodialysis method. RESULTS The microdialysis recoveries of matrine were inversely proportional to the perfusion rate, independent of matrine concentration. The positive dialysis recoveries in vitro were similar to the retrodialysis recoveries, while the in vitro recoveries were greater than those in vivo. The in vivo recoveries of concentric cannula probe dialysis were greater than those of linear probe dialysis. CONCLUSION The in vitro positive dialysis method is simple and accurate, which can be used for matrine pharmacokinetic study.
