摘  要:目的:观察糖脂清颗粒对2型糖尿病模型大鼠血糖、学习记忆能力及IRS-1在海马组织表达的影响并探讨其作用机理。方法:采用高糖高脂饮食加小剂量腹腔注射链脲佐菌素建立2型糖尿病大鼠模型,将成模大鼠按随机数字表法分为模型组、吡格列酮组、糖脂清高剂量组、糖脂清中剂量组,另设正常组。给药4周后观察大鼠血糖、Morris水迷宫测试、IRS-1在各组大鼠海马部位的表达。结果:1血糖:糖脂清颗粒可以显著改善2型糖尿病模型大鼠血糖水平,并以高剂量组效果最显著(P<0.01);2空腹血清胰岛素:糖脂清高剂量组、糖脂清中剂量组及吡格列酮组大鼠治疗后空腹血清胰岛素值高于治疗前(P<0.05),糖脂清高剂量组、糖脂清中剂量组及吡格列酮组空腹血清胰岛素值高于糖尿病模型组(P<0.05);3水迷宫测试:糖脂清中剂量组、糖脂清高剂量组大鼠穿越平台次数均多于模型组,差异有统计学意义(P<0.01或P<0.05);糖脂清高剂量组大鼠穿越原平台象限游泳时间/总时间高于模型组(P<0.05);4免疫组织化学染色:糖脂清高剂量组、糖脂清中剂量组大鼠海马组织IRS-1水平较模型组高(P<0.01或P<0.05)。结论:糖脂清颗粒可以显著降低高糖高脂饮食加小剂量链脲佐菌素诱导的2型糖尿病大鼠模型的血糖水平,具有促进大鼠学习记忆能力的作用,其作用机理可能与改善糖代谢、提高IRS-1在海马组织的表达、修复脑内胰岛素信号传导通路有关。Objectives:To observe TangZhiQing Granules effect on blood glucose, ability of learning and memory, and IRS-1 expression in the hippocampus in type 2 diabetic model rats and exploring its mechanism. Methods:Use high sugar and high fat diet combined with low-dose Streptozotocin to establish rat model of type 2 diabetes. Type 2 diabetic model rats were randomly assigned to model groups, pioglitazone group, T high-dose group, T middle-dose group. In addition set a normal group. After 4 weeks of medication, observed in rats following indicators: ① glucose; ②Morris water maze test; ③ IRS-1 expression in hippocampus of rats. Results: ①Blood glucose: TangZhiQing Granules can significantly reduce blood sugar levels of rats of type 2 diabetes. T high-dose group effects most notably (P 〈 0. 01 ).②fasting serum insulin: fasting serum insulin of T high-dose group, T middle-dose group, and pioglitazone group after treatment are higher than before treatment ( P 〈 0. 05 ) ; after treatment fasting serum insulin of T high-dose group, T middle-dose group, and pioglitazone group are higher than model groups ( P 〈 0. 05 ). ③ Water maze test : the number of T middle-dose group and T high-dose group across platform is more than model group, there was a significant difference (P 〈 0.01 or P 〈 0. 05 ); T high-dose group cross platform-quadrant swimming time/total time higher than the model group (P 〈 0.05 ). ④ Immunohistochemical staining: T high-dose group and T middle-dose group IRS-1 levels in rat hippocampus higher than model group(P 〈 0. 01 or P 〈 0. 05 ). Conclusion: TangZhiQing Granules can significantly reduce blood sugar levels of the high sugar, high fat diet and low-dose Streptozotocin-induced model rats of type 2 diabetes. TangZhiQing Granules has the effect of promoting the learning and memory ability of rats.
