摘 要:目的:运用荧光免疫组织化学技术揭示刮痧对局部组织中化学成分的影响。方法:以大鼠为实验对象,先剔除背毛暴露出脊柱两侧皮肤,然后用刮痧板沿一侧相当于人体膀胱经走行的部位由上向下刮拭。出痧后将动物灌流固定,并取下出痧部位的皮肤组织,同时取下正常大鼠的相应部位用于对照。取下的组织放在25%高渗糖中脱水后用冰冻切片机制成20μm的系列组织片,然后分别用于神经元型一氧化氮合酶(n NOS)和组胺(HA)抗体进行荧光免疫组织化学染色。结果:在刮痧后的组织中n NOS和HA阳性表达明显增强,n NOS标记主要分布在表皮的细胞间隙,HA标记集中分布在表皮层与真皮层之间,与正常组织比较差异有统计学意义。结论:刮痧对局部组织中n NOS和HA的表达均起到上调作用,提示荧光免疫组织化学技术可能成为揭示刮痧生物学效应的有效方法之一。Objective: To investigate the influence of guasha (scraping therapy) on the chemical expression of local tissue by using fluorescent immunohistochemieal technique. Methods :The rat was used in this experiment. The skin on both sides of spinal column was exposed by shearing wool from the back of rats. Subsequently, guasha was earried out on the area along a side of spinal column from the front to the rear, the corresponding site of urinary bladder meridian in the human body. After shazhen (subcutaneous hemorrhage showed in scattered red spots on the skin) appeared, the local tissues of shazhen and similar areas of normal rats were dissected out following perfusion with 4% paraformaldehyde, and put into 25% sucrose solution. Serial tissue sections were cut at a thickness of 20 mm on a cryostat. The chemical expression of tissue was demonstrated by using fluorescent immunohistochemical staining for neuronal nitrogen monoxide synthetase (nNOS)and histamine (HA). Results The positive labeling of nNOS and HA increased in the local tissue after guasha, in which nNOS-labeling mainly distributed between the cells in the epidermis and HA-labeling concentrated between the epidermis and the dermis. The morphology of the tissues with shazhen was significantly different from that of the tissue in the control. Conclusion :These results indicate that guasha plays a role in up-regulating the level of nNOS and HA in the local tissue, and suggest that fluorescent immunohistochemical technique may be one of the effective means to understand the biological effectiveness of guasha.
