摘  要:目的观察去势大鼠阴茎海绵体胱硫醚γ裂解酶(Cystathionine-γ-lyase,CSE)/硫化氢(Hydrogen sulphide,H2S)的变化,进一步探讨勃起功能障碍的发病机制。方法雄性SD大鼠72只分4组:对照组、假手术组,去势组和去势炔丙基甘氨酸(PAG,CSE阻断剂)组,检测基础条件下和阿扑吗啡(Apomorphine,APO)刺激后的海绵体内压(Intracavernous pressure,ICP)及勃起率;激光共聚焦显微镜检测CSE在大鼠勃起不同时期阴茎海绵体组织中的表达,敏感硫电极测定H2S在勃起不同时期的含量。结果与假手术组比较,去势组和去势PAG组ICP与勃起率下降(P<0.01);且去势PAG组较去势组ICP明显下降(P<0.01);阿朴吗啡刺激后,与假手术组比较,勃起前去势组和去势PAG组CSE蛋白表达降低(P<0.01),勃起中去势各组较假手术组CSE蛋白表达降低(P<0.01),且去势PAG组较去势组CSE蛋白表达明显降低(P<0.01);勃起后各组间CSE蛋白表达变化无差异。勃起前和勃起中去势各组较假手术组H2S含量下降(P<0.05),且勃起中去势PAG组较去势组H2S含量明显下降(P<0.01):勃起后去势组和去势PAG组较假手术组H2S含量下降(P<0.01)。结论去势大鼠勃起功能障碍与CSE和H2S表达下降有关。Objective To explore changes of CSE / H2 S in the corpus cavernosum of castration rat in order to further study the pathogenesis of erectile dysfunction. Methods 72 male SD rats were selected and randomly divided into four groups: the normal group,sham operation group,castration group and castration + PAG group. The ICP and ratio of penile erection under basic condition and APO induction conditions were detected. The expression of CSE in the corpus cavernosum during different periods of penile erection was detected by LSCM,and the H2 S during different periods of penile erection was detected by sensitive sulphur electrode method. Results The ICP and the ratio of penile erection decreased significantly in castration group and castration + PAG group compared with sham operation group( P〈0. 01); and the expression of ICP in castration group obviously decreased compared with that in castration + PAG group( P〈0. 01). Moreover,compared with sham operation group,the expression of CSE protein significantly decreased in castration group and castration + PAG group before erection stimulation by APO( P〈0. 01). During the erection,the expression of CSE protein in each castration group decreased significantly compared with sham operation group( P〈0. 01); the expression of CSE protein in castration + PAG groups decreased significantly compared with the castration group( P〈0. 01). After erection,there were no significant difference in the expression of CSE protein between different groups. Before and during erection,the level of H2 S in all castration groups decreased significantly compared with sham operation group( P〈0. 05),and the level of H2 S in castration + PAG group decreased significantly compared with castration group( P〈0. 01). After erection,the level of H2 S in castration group and castration + PAG groups decreased compared with that in sham operation group( P〈0. 01).
