摘 要:目的研究具药用价值的海洋微藻杜氏盐藻Dunaliella salina和亚心形扁藻Platymonas subcordiformis在不同培养容器以及光照下的细胞生长情况,为促进海洋微藻生物资源的培养保存提供参考。方法选取杜氏盐藻和亚心形扁藻2种海洋微藻为生物材料,在植物培养箱中分别利用三角瓶和试管,在正常光照和遮光光照条件下开展为期10 d的一次性培养试验,利用倒置显微镜和血球计数板观测微藻细胞密度,统计分析不同培养容器和光照对2种海洋微藻生长影响的作用。结果在该实验条件下,2种海洋微藻的细胞生长均表现出初期生长缓慢、中期生长快速、后期生长放缓的趋势。杜氏盐藻在三角瓶中正常光照培养条件下的生长最好(504×104个/m L),其次为在试管中正常光照培养的条件下(364×104个/m L);亚心型扁藻细胞在三角瓶中正常光照培养的条件下生长最好(227×104个/m L),其次为在三角瓶中遮光处理的条件下(177×104个/m L)。结论培养容器(三角瓶和试管)和光照(正常光照和遮光)对2种海洋微藻生长的影响均具有统计学意义,在进行微藻生物资源培养和保存时,应充分考虑或利用这2种因素的作用。Objective To investigate the cell growth of two marine microalgae Dunaliella salina and Platymonas subcordiformis in different containers and light conditions,hence to provide reference for marine microalgal cultivation and preservation.Methods D.salina and P.subcordiformis were cultured in a plant growth chamber in sea water enriched with t/2 medium for 10 days using a batch culture.Conical flasks and tubes were chosen as containers for cell cultivation; normal light and shading light were set up.Cell density was monitored using a hemacytometer under microscopy to investigate the effects of container and light on the cell growth of two marine microalgae.Results The two microalgae showed a growth tendency of slowly growing in the early stage,rapidly growing in the middle stage and steadily growing in the late stage.D.salina had the highest cell densities (504 × 10^4 cells/mL) when cultivated in flask with normal light,followed by in the tube and normal light condition (364× 10^4 cells/mL).P.subcordiformis showed the highest cell densities (227× 10^4 cells/mL) in the flask with normal light,followed by in the flask with shading light (177× 10^4 cells/mL).Conclusion Containers (Conical flasks and tubes) and light conditions (normal light and shading light) show significant effects on the growth of the two marine microalgae D.salinaand P.subcordiformis.The differences in container and light should be fully considered while cultivating and preserving microalgal resources.
