摘 要:目的:建立加味玉屏风颗粒中黄芪甲苷的含量测定方法。方法:采用高效液相色谱法,色谱柱为Diamonsil C18柱(4.6mm×250mm);流动相为乙腈-水(34:66);ELSD参数:漂移管温度105℃;载气流量2.5L/min。结果:黄芪甲苷进样量在4.821μg^24.104μg范围内与峰面积线性关系良好,回归方程为:A=1.3315C+4.7412,r=0.9992,平均回收率为99.52%,(RSD=2.42%,n=6)。结论:该方法简便、准确、重现性好,可用于加味玉屏风颗粒的质量控制。ObjectiveTo establish a method of determining the astragaloside content in Jiawei Yupingfeng granules by HPLC- evaporating light scattering detector(ELSD). Methods The HPLC- ELSD was used in the quantitative analysis by using the Diamonsil C18 chromatography column(250 mm × 4. 6 mm,5 μm) and acetonitril- water(34 ∶ 66) as the mobile phase. The flow rate was 1. 0 mL /min. The tube temperature of the drift tube was 105℃. The flow rate of carrier air was 2.5 L /min. Results The linear range of astragaloside reference substance was 4. 821μg - 24. 104μg,the regression equation was A = 1. 3315 C + 4. 7412,r = 0. 9992. The average recovery rate was 99. 52%,RSD was 2. 42% %. Conclusion The method is strongly specific,simple to operate and accurate in the detection results,which can be used as the determination method of the astragaloside content in Jiawei yupingfeng granules.
