摘  要:为比较不同核酸提取方法检测禽流感病毒敏感性的差异,将 H5N1禽流感抗原做10-2-10-10稀释,分别用 Trizol 法、磁珠法和膜吸附法提取上述抗原稀释度核酸模板,采用荧光 RT-PCR 对其进行评价。结果表明,在不同稀释度的抗原的检测中,3种核酸提取方法以膜吸附法提取效率最高,其依次为磁珠法、Trizol 法,经方差分析,3种核酸提取方法之间存在显著性差异(P<0.05)。说明 3种核酸提取方法提取效率存在显著差异,核酸提取效率的高低影响禽流感病毒荧光 RT-PCR 检测的敏感性,建议实验室根据实际情况选择核酸提取的方法。To compare the efficiency of different nucleic acid extraction methods for detection of avian influ-enza virus.H5N1 avian influenza virus was used as template,Trizol,magnetic beads and membrane ad-sorption methods were used for nucleic acid extraction,H5N1 avian influenza virus were diluted from 10 -2 to 10-10 ,the efficiency of the three methods were evaluated through real-time RT-PCR.The variance anal-ysis showed there were significance differences among the three different methods(P〈0.05).The mem-brane adsorption extraction method is the most efficient one,and followed by magnetic beads and Trizol extraction.The nucleic acid extraction efficiency is closely related to the sensitivity of the real-time RT-PCR results.The results suggested that the appropriate detection methods should be chosen for detection of clinical specimens based on actual situation.
