摘 要:目的研究S100A4siRNA对关节炎大鼠炎症及细胞因子肿瘤坏死因子α(TNF-α)、IL-1β和血管内皮生长因子(VEGF)表达的影响。方法建立大鼠佐剂性关节炎模型,造模后第11天给模型干扰组大鼠关节腔内注射S100A4siRNA片段。观察各组大鼠关节炎指数(AI)的变化及踝关节的病理变化;ELISA检测血清TNF-α、IL-1β和VEGF的表达变化。结果模型干扰组大鼠TNF-α、IL-1β和VEGF表达水平均比模型组降低(P<0.05);AI值及踝关节病理积分与模型组比较显著下降,差异有统计学意义(P<0.05)。结论抑制S100A4基因表达,能显著降低致炎细胞因子TNF-α、IL-1β及促血管生成因子VEGF的表达,减轻关节滑膜的病理损伤。Objective To observe the effects of S100A4 siRNA on the expression of serum TNF‐α,IL‐1βand VEGF in adjuvant arthritis rats .Methods Adjuvant arthritis rat models were established and were randomly divided into model group and interfere group .On Day 11 ,rats in interfere group were injected with S100A4 siRNA fragment in articular cavity .Arthritis index (AI) chan‐ges and pathological changes of ankle joint were observed .The levels of serum TNF‐α and IL‐1β ,VEGF were detected by ELISA . Results Compared with that of model group ,the levels of serum TNF‐ α ,IL‐1β and VEGF were reduced significantly in interfere group (P〈 0 .05) ;variances of AI and pathological scores in interfere group were diminished significantly (P〈 0 .05) .Conclusion Inhibition of the expression of S100A4 gene can significantly reduce the expression of inflammatory factor TNF‐α ,IL‐1 β and angio‐genesis factor VEGF ,and improve the pathological injury of synovial membrane .
