摘 要:目的对两种不同脱蛋白方法处理后的新型牛松质骨移植替代材料进行理化性质的检测和分析,探讨更适合临床应用的新型骨移植替代材料。方法采用游标卡尺对新型牛松质骨移植替代材料的尺寸进行测量,通过电镜技术进行细微结构的观察,并同时测量标本的孔径和孔隙率。通过X射线衍射实验完成材料中羟基磷灰石含量的检测,采用电感耦合等离子体原子发射光谱法测定钙、磷含量,并据此计算钙、磷原子比。同时按凯氏定氮法测定材料中蛋白质的含量,索氏提抽法测定材料中脂肪的含量。并分别按推荐性国家标准(GB/T)中规定的方法对材料中的砷、镉、汞和铅进行测量。结果经过氧化氢法及胃蛋白酶法对牛松质骨脱蛋白后的骨移植替代材料具有相似的尺寸,较满意的孔径大小与孔隙率,在X射线衍射实验中都只有一个物相即Ca5(PO4)3(OH)相,主要成分为羟基磷灰石。两种脱蛋白方法处理后的骨移植替代材料中脂肪含量、蛋白质含量及重金属含量较脱蛋白处理前明显降低(F脂肪=85.727,P<0.01;F蛋白质=227.963,P<0.01;t铅=14.494,P<0.01;F镉=496.486,P<0.01;F砷=1741.374,P<0.01;F汞=84.592,P<0.01)。较过氧化氢法,胃蛋白酶法处理的牛松质骨骨移植替代材料中蛋白质及重金属含量更低,钙、磷原子比与人骨更加接近。结论过氧化氢脱蛋白法及胃蛋白酶脱蛋白法处理的两种牛松质骨移植替代材料的理化性质较处理前有明显改善,但经胃蛋白酶脱蛋白法的牛松质骨移植替代材料理化性质更佳。[Objective] To test and analyze the physical and chemical properties of the new bovine cancellous bone graft substitute materials deproteinized by two different methods and to discuss which one is more suitable for the clinical application. [ Method ] The vernier caliper was used to measure the sizes of the new bovine cancellous bone graft substitute materials; the electron technology was used to finish its subtle institutions observation, and mea- sure the aperture and porosity of the specimens at the same time. The X-ray diffraction experiment was used to in- spect completed hydroxyapatite content of materials, the inductively coupled plasma atomic emission spectrometry was used to test calcium, phosphorus content, and calculate the calcium/phosphorus atoms ratio. The protein content of materials was tested by kieldahl method and the fat content by soxhlet extration method. Arsenic, cadmium, mercury, lead of materials was measured separately by the specified methods according to recommended national stan- dards (GB/T) at the same time. [Results] The bone graft substitute materials which were the bovine cancellous bones deproteinized by hydrogen peroxide and pepsin methods were similar in appearance, size, satisfied aperture and porosity, and both existed only one phase that was Ca_5(PO_4)_3(OH) by X-ray diffraction, and the main composition was hydroxyapatite.The protein,fat and heavy metal content of bone graft substitute materials were deproteinized by two different methods, the protein, fat and heavy metal content was significantly lower than pretreatment (F_fat=85.727, P 〈0.01; F_protein=227.963, P 〈0.01; t_Pb=14.494, P 〈0.01; F_Cd=496.486, P 〈0.01; F_As=1741.374, P 〈0.01; F_Hg=84.592, P 〈0.01). Compared with hydrogen peroxide method, the bone graft substitute materials deproteinized by pepsin method were lower in content of protein and the heavy metal, calcium/phosphorus atoms ratio was closer to human bone.
