摘  要:目的:观察巴曲酶对实验性自身免疫性脑脊髓炎(EAE)小鼠发病情况及脑组织纤维蛋白沉积的影响。方法:将50只雌性C57BL/6小鼠随机分为正常对照组、EAE对照组、巴曲酶低剂量治疗组、中剂量治疗组、高剂量治疗组,采用MOG35-55免疫C57BL/6小鼠诱发EAE动物模型。观察小鼠发病潜伏期、进展期和高峰期神经功能障碍评分变化;免疫组化染色法检测脑组织纤维蛋白沉积,计算其相对阳性面积。结果:与EAE对照组比较,巴曲酶各治疗组的潜伏期明显延长,进展期明显缩短,发病高峰期神经功能障碍评分明显降低,呈剂量依赖关系;巴曲酶各治疗组中纤维蛋白沉积量较EAE组明显减少,且呈剂量依赖关系;EAE对照组、巴曲酶各治疗组的纤维蛋白沉积量与潜伏期呈明显负相关,与进展期及发病高峰期神经功能障碍评分呈明显正相关。结论:巴曲酶通过减少脑组织纤维蛋白沉积而发挥对EAE的保护作用。Objective:To explore the effect of batroxobin on fibrin deposition in C57BL/6 mice with experimental allergic encephalomyelitis(EAE). Methods:50 female C57BL/6 mice were randomly divided into normal group, EAE control group, low, median, and high dose batroxobin treatment groups. C57BL/6 mice was immunized with MOG 35-55 to establish the EAE model. The latency and progress of EAE were observed; the nervous dysfunction was scored at the peak of EAE.Immunohistochemical method was used to detect fibrin deposition. Results: Compared with the EAE control group, the latent period of each batroxobin treatment group was extended apparently; the progression period was shorten obviously; the nervous dysfunction score was decreased significantly in dose-dependent relationship. The fibrin deposition in each batroxobin treatment group was dose-related and much less than that in the EAE control group. The immunohistochemical positive area of EAE control group and each batroxobin treatment group was negatively correlated with the latent period, positively correlated with the progression period and the neurodys-function scores. Conclusion: Batroxobin shows its protective effect on EAE through reducing fibrin deposition.
