摘  要:对转人源化Fat-1基因猪的15头传代仔猪进行PCR检测,发现有7头阳性猪,其断奶后成活5头仔猪。对这5头仔猪进行Southern印迹杂交试验,确认3头仔猪整合有hFat-1基因。采用荧光实时定量PCR法测定原代转hFat-1基因公猪及3头阳性仔猪外源基因的拷贝数,分别是2、1、1、2个拷贝。转hFat-1基因猪能够将外源基因遗传到下代仔猪,但外源基因具有遗传不稳定的趋向。Fifteen heads of hFat-1 transgenic offspring piglets were detected by PCR, 7 heads of piglets were positive, and 5 heads of piglets survived after weaning.Southern blotting cross experiment confirmed that the exogenous gene hFat-1 had been integrated into 3 piglets among these 5 piglets.The copy numbers of hFat-1 in primary hFat-1 transgenic boar and 3 positive piglets were detected by fluorescence real-time Q-PCR, and they were 2, 1, 1, and 2 copies respectively.The integration of exogenous gene in hFat-1 transgenic pigs could be inherited to F1 -generation piglets, but could not be stably inherited.
