摘  要:基于亲水相互作用色谱(HILIC)技术,建立了可用于茎直黄芪中苦马豆素测定方法。样品用甲醇提取,挥干后用0.05 M盐酸溶解,氯仿和水饱和正丁醇依次萃取。Waters Oasis MCX柱净化,Waters XBridge HILIC色谱柱分离,流动相为20 mmol/L磷酸二氢铵溶液(pH=7.8)-乙腈(20∶80,V/V),检测波长为205 nm,流速为0.5 mL/min。结果表明,应用该方法测定了茎直黄芪中苦马豆素含量,为46.44 mg/kg。建立的高效液相色谱检测方法简单、快捷、准确度高。Swainsonine in Aaragalus grictus was detected by using hydrophilic interaction liquid chromatography (HILIC).The sample was extracted by methanol in under ultrasonic assistance conditions.The extract was evaporated to dryness and redissolved by 0.05M hydrochloric acid and extracted by chloroform and n-butyl alcohol and cleaned up on a Waters Oasis MCX solid phase extraction cclumn.The separation was carried out by using Waters XBridge HILIC and 20 mmol/L NH4H2PO4(pH =7.8)-acetonitrile (20∶ 80,v/v) as mobile phase.The detection was set at 205 nm and the flow rate was 0.5 mL/min.The results showed that,by using this method,the content of SW in A.strictus was determined.the content of SW in A.strictus was 46.44 mg/kg.The conclusion was drawn that the method was simple and accurate,which could be used to determine SW content in A.strictus.
