摘  要:目的观察脑通汤对血管性痴呆大鼠行为学、海马CA1区神经元凋亡基因Bcl-2、Bax表达及Bcl-2/Bax比例变化的影响。方法采用改良四血管法(4-VO)制备血管性痴呆(VD)模型,Morris水迷宫测定大鼠学习、记忆能力,免疫组化法检测海马CA1区神经元凋亡基因Bcl-2、Bax蛋白表达水平,计算Bcl-2/Bax比例。结果与模型组比较,脑通汤大、中剂量组逃避潜伏期缩短,穿越原平台次数增多,Bax表达降低,Bcl-2表达增高,Bcl-2/Bax比例升高。结论脑通汤可明显提高Bcl-2表达,减少Bax表达,升高Bcl-2/Bax比例,抑制神经细胞凋亡,从而减轻脑缺血对神经血管单元的损伤,改善VD大鼠学习、记忆能力,这可能是其治疗血管性痴呆的作用机制之一。Objective To observe the effect of Naotong Decoction on the learning and memory abilities,and the neuronal apoptosis genes Bax,Bc1-2 expression and Bcl-2/Bax ratio in CA1 region of the hippocampus on vascular dementia(VD) rats. Methods The vascular dementia models were established by modified Pulsinelli 4 vascular occlusions method(4VO). Morris water maze was used to test learning and memory abilities of rats. Immunohistochemistry was used to detect the neuronal apoptosis gene Bcl-2,Bax expression in CA1 region of the hippocampus,and to calculate Bcl-2/Bax ratio. Results Compared to the model group,the escape time was shortened and times of spanning the previous platform increased in Naotong Decoction large-dose and middle-dose treatment group. Meanwhile,the expression of Bax protein was reduced and the expression of Bcl-2 protein was promoted,the ratio of Bcl-2/Bax rised. Conclusion Naotong Decoction can obviously improve the expression of Bcl-2,reduce the expression of Bax,increases the Bcl-2/Bax proportion,inhibit of nerve cells apoptosis,thus to reduce the damage to nerve vascular unit,to make it improve learning and memory ability of VD rats. This may be one of the mechanisms of Naotong decoction in the treatment of vascular dementia.
