栀子柏皮汤合煎液与分煎液中有效成分含量的比较研究

摘　　要：目的探究栀子柏皮汤合煎与分煎提取液中有效成分栀子苷、西红花苷-Ⅰ、西红花苷-Ⅱ、盐酸药根碱、盐酸巴马汀、盐酸小檗碱、甘草苷和甘草酸的变化。方法采用Agilent TC-C18(4.6mm×250mm,5μm)色谱柱,流动相以乙腈-0.2%磷酸水溶液,分别在238nm波长下检测栀子苷、甘草苷和甘草酸,在265nm波长下检测盐酸药根碱、盐酸巴马汀和盐酸小檗碱,在440nm波长下检测西红花苷-Ι和西红花苷-ΙΙ;流速为0.8 ml/min,梯度洗脱,柱温30℃。结果合煎剂中总生物碱、甘草酸的含量低于分煎剂;西红花苷类化合物(西红花苷-I、西红花苷-ΙΙ)的含量明显高于分煎剂;合煎剂中栀子苷、甘草苷与分煎剂中含量无明显变化。结论栀子柏皮汤合煎剂中有效成分溶出增加、毒性成分含量降低,比分煎剂质量优越。Objective To explore dissolution Changes of geniposide,liquiritin,crocin-1,jatrorrhizine hydrochloride,crocin-2、berberine hydrochloride、palmatine hydrochloride and glycyrrhizic acid in the combined decoction and the separated decoction of Zhizi Baipi Decoction simultaneously. Methods The HPLC analysis was performed on an Agilent TC-C18column（4. 6 mm × 250 mm,5μm） column,the column temperature was 30℃; acetonitrile-0. 2% phosphoric acid as the mobile phase,0. 8 μml/min gradient elution; detection wavelengths were 238nm（geniposide,liquiritin and glycyrrhizic acid）,440nm（crocin-1,crocin-2）and 265nm（jatrorrhizine hydrochloride,berberine hydrochloride and palmatine hydrochloride）. Results The contents of total alkaloids and glycyrrhizic acid in the combined decoction were lower than that in the separated decoction. The contents of crocin-1and crocin-2 in the combined decoction were higher than that in the separated decoction. The contents of geniposide and liquiritin in the combined decoction were almost as same as that in the separated decoction. Conclusion The Combined Decoction of Zhizi Baipi Decoction has more dissolution of active ingredients and lower toxic side effects than the Separated Decoction of Zhizi Baipi Decoction.
