摘 要:目的评价扶正除疫颗粒抗甲型H1N1流感病毒感染的效果。方法将112只小鼠随机分为正常对照组、病毒模型组、达菲对照组、扶正除疫组,每组28只。正常对照组和病毒模型组以生理盐水灌胃,每次0.4 ml/只;达菲对照组以达菲水溶液0.025 g/(kg·d)灌胃,扶正除疫组以扶正除疫颗粒水溶液12 g/(kg·d)灌胃,均每日1次,共连续灌胃7天。给药第3天,除正常对照组外其余3组以H1N1亚型季节性流感病毒鼠肺适应株FM1-6-E2滴鼻造模,攻毒剂量为5LD50,50μl/只,造模后1h各组继续给药。从攻毒后感染之日起,每组取10只,连续观察14天,观察小鼠死亡情况;各组余18只于攻毒后3天、6天、9天计算肺指数,检测病毒滴度,观察肺组织病理变化。结果病毒模型组10只小鼠全部死亡;达菲对照组和扶正除疫组死亡率均为0。攻毒后第9天,达菲对照组、扶正除疫组与病毒模型组比较,肺指数明显降低(P<0.05或P<0.01);攻毒后第3天、6天,达菲对照组、扶正除疫组与病毒模型组比较,肺组织病毒滴定度明显降低(P<0.05或P<0.01)。达菲对照组与扶正除疫组小鼠肺脏病理变化较为轻微,炎症反应程度明显弱于病毒模型组。结论扶正除疫颗粒对小鼠感染甲型H1N1流感病毒后具有死亡保护作用,对流感病毒在宿主体内复制有抑制作用。Objective To evaluate the efficacy of Fuzheng Chuyi Granules against the infection of influenza Avirus HI N1. Methods We randomly divided 112 mice into normal control group, virus model group, tamiflu control group and Fuzheng Chuyi group, 28 rats in each group. Once per day for 7 days, normal control group and virus model group was given normal saline O. 4 ml each, tamiflu control group was given tamiflu's aqueous solution O. 025 g/ (kg · d) and Fuzheng Chuyi group Fuzheng Chuyi Granules' aqueous solution 12 g/ (kg · d) all by intragastric administration. On day 3, all groups except normal control group was induced by nose dripping HI N1 seasonal influenza virus mice lung adapted strain FM1-6-E2 (infection dosage 5LDs0, 50 μ1 each) and continued medication 1 hour after modeling. After infection day, took l0 mice out of each group to observe the mortality for 14 days. The rest 18 mice were calculated on lung index, tested for virus titer and observed on pathological changes of lung tissue on day 3, 6, 9 after infection. Results 10 mice in virus model group all died, while 10 mice in tamiflu control group and in Fuzheng Chuyi group didn't die. On day 9 after infection, comparing tamiflu control group, Fuzheng Chuyi group with virus model group, lung index was significantly lower (P 〈 0.05 or P 〈 0. 01 ). On day 3, 6 after infection, comparing tamiflu control group, Fuzheng Chuyi group with virus model group, virus titer of lung tissue was obviously lower ( P 〈0.05 or P 〈0. 01 ). Pathological changes of lung tissue in tamiflu control group and Fuzheng Chuyi group were minor and degree of inflammation was notedly less severe than that of virus model group. Conclusion Fuzheng Chuyi Granules has protective effect against mice death after infection of influenza A virus HI N1 and sup-pressed influenza virus's replication in the host.
