佛手益气活血汤对血栓闭塞性脉管炎模型大鼠TXB_2、6-Keto-PGF_(1)α、VEGF表达的影响

摘　　要：目的:通过观察佛手益气活血汤对血栓闭塞性脉管炎(TAO)模型大鼠血栓素B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α)、血管内皮生长因子(VEGF)表达的影响,探讨佛手益气活血汤对大鼠TAO的治疗作用及其机制。方法:将Wistar大鼠60只随机分为假手术组,模型对照组,佛手益气活血汤高、中、低剂量组,每组12只,假手术组及模型对照组大鼠分别灌胃10 m L/kg生理盐水,佛手益气活血汤高、中、低剂量组大鼠分别灌胃佛手益气活血汤30.0、15.0、7.5 g/kg;连续灌胃14天后检测大鼠血浆TXB2、6-Keto-PGF1α及VEGF表达水平。结果:佛手益气活血汤对模型大鼠血浆6-Keto-PGF1α水平具有升高作用,对TXB2及肌肉VEGF表达具有降低作用,与模型对照组比较差异具有统计学意义(P<0.05)。结论:佛手益气活血汤对血栓闭塞性脉管炎模型大鼠病变程度具有一定的改善作用,可能通过调节血浆TXB2及6-K-PGF1α平衡,降低组织VEGF表达而发挥作用。Objective： To explore curative effects of Fo Shou Yi Qi Huo Xue decoction in treating rats with thromboangiitis obliterans（TAO） and its mechanism by observing its influence on TXB2, 6-Keto-PGF1α and VEGF of TAO rats. Methods： Sixty rats were randomly divided into sham operation group, model control group, high, moderate and low dose groups of Fo Shou Yi Qi Huo Xue decoction, 12 rats each group, sham operation group and model control group accepted intragastric administration of physiological saline, 10 m L/kg, the rats in high, moderate and low dose groups of Fo Shou Yi Qi Huo Xue decoction were perfused with Fo Shou Yi Qi Huo Xue decoction at the concentrations of 30.0 g/kg, 15.0 g/kg and 7.5 g/kg respectively; the expressions of TXB2, 6-Keto-PGF1α and VEGF of TAO rats were detected after perfused for 14 days consecutively. Results： Fo Shou Yi Qi Huo Xue decoction demonstrated the functions of raising the level of 6-Keto-PGF1α, lowering the expressions of TXB2 and VEGF the difference had statistical meaning compared with model control group（P〈0.05）. Conclusion： Fo Shou Yi Qi Huo Xue decoctio n could improve TAO rats to a certain degree, it works by regulating the balance between TXB2 and 6-Keto-PGF1α in the plasma and decrease the expressions of VEGF in the tissue.
