摘 要:目的探讨三维适形调强放疗(intensity modulated radiation therapy,IMRT)的放射线对鼠骨细粒棘球蚴生发细胞生长及凋亡的影响。方法骨细粒棘球蚴病子午沙鼠20只,将骨细粒棘球蚴内囊中的生发细胞进行分离,并进行体外培养,对体外培养成功的骨细粒棘球蚴生发细胞分为IMRT放疗组和空白对照组。IMRT放疗组给予总剂量为40Gy的IMRT放疗,对照组仅给予假照射。于放疗结束24h后采用流式细胞仪检测骨细粒棘球蚴生发细胞凋亡率,采用Western blot法检测骨棘球蚴生发细胞中凋亡相关蛋白caspase-3蛋白表达情况,采用RT-PCR法定量检测骨棘球蚴生发细胞caspase-3mRNA表达水平。结果 IMRT作用24h后,IMRT放疗组骨细粒棘球蚴生发细胞凋亡率、caspase-3蛋白及mRNA表达灰度值((67.21±14.40)%、7.761±1.447、59.44±19.28)明显高于对照组((2.91±0.51)%、0.129±0.054、1.17±0.50),差异均有统计学意义(P<0.05)。结论 IMRT对骨细粒棘球蚴生发细胞生长有明显诱导凋亡作用,其作用机制可能是通过调节凋亡相关蛋白caspase-3而实现的。Objective To observe the influence of intensity modulated radiation therapy(IMRT)on the growth and apoptosis of germinal cells of bone echinococcosis in Meriones meridianus.Methods Twenty Meriones meridianus with bone echinococcosis were separated the germinal cells of bone echinococcosis and cultured in vitro.The successfully cultured of germinal cells of bone echinococcosis were divided into IMRT group and blank control group.IMRT group received IMRT at the total dose of 40 Gy,and blank control group received no IMRT.The apoptosis rate of germinal cells was detected by flow cytometer,and the expressions of caspase-3protein and mRNA were detected by Western blot and RT-PCR 24 hours after radiation.Results The apoptosis rate,and the expressions of caspase-3protein and mRNA in germinal cells were significantly higher in IMRT group((67.21±14.40)%,7.761±1.447,59.44±19.28)than those in blank control group((2.91±0.51)%,0.129±0.054,1.17±0.50)(P〈0.05).Conclusion It is evident that the IMRT can promote the apoptosis of germinal cells in Meriones meridianus by regulating the expression of caspase-3.
