摘 要:目的探讨三维适形调强放疗(intensity modulated radiationtherapy,IMRT)对子午沙鼠骨细粒棘球蚴原头节的杀灭作用及其对葡萄糖-6-磷酸酶表达的抑制作用。方法双股骨骨细粒棘球蚴病子午沙鼠40只,左侧股骨骨细粒棘球蚴病区给予总剂量为40Gy的IMRT放疗,右侧股骨骨细粒棘球蚴病区仅给予假照射作为阴性对照。于放疗结束1周后取鼠左、右侧股骨细粒棘球蚴原头节行伊红染色检测原头节坏死率,采用硝酸铅法检测葡萄糖-6-磷酸酶的活性变化。结果 IMRT作用1周后,可见子午沙鼠左侧股骨骨细粒棘球蚴原头节整体结构消失,出现深蓝着色;左侧股骨骨细粒棘球蚴原头节坏死率((71.26±21.85)%)高于右侧((5.93±5.77)%),差异有统计学意义(P<0.05);子午沙鼠左侧股骨骨细粒棘球蚴原头节的葡萄糖-6-磷酸酶活性较右侧明显减弱,葡萄糖-6-磷酸酶产物光密度值(0.271 2±0.104 2)明显低于右侧(0.642 9±0.398 5),差异有统计学意义(P<0.05)。结论 IMRT可促进鼠骨细粒棘球蚴原头节的死亡,其作用机制可能是通过改变葡萄糖-6-磷酸酶的活性而实现。Objective To observe the influence of intensity modulated radiation therapy(IMRT)on killing protoscolex of bone echinococcosis in Meriones meridianus and inhibiting the expression of glucose-6-phosphatase(G-6-P).Methods Forty Meriones meridianus with bone echinococcosis received IMRT at the total dose of 40 Gy on lesions of the left femur,and the right femur received no IMRT.One week after IMRT,the left and right samples of bone echinococcosis were taken out to observe the necrosis rate by the Hematoxylin-Eosin staining and the activity of G-6-P by Lead nitrate method.Results One week later,the overall structure of protoscolex of bone echinococcosis of left femur disappeared,showing dark blue color.The necrosis rate was(71.26±21.85)% of the left side,higher than that of the right side((5.93±5.77)%)(P〈0.05).The activity of G-6-P was weaker of the left side than that of the right side,and the optical density value of succinate dehydrogenase was 0.271 2±0.104 2of the left side,lower than that of the right side(0.642 9±0.398 5)(P〈0.05).Conclusion IMRT can promote the echinococcosis apoptosis in the Meriones meridianus by changing the activity of G-6-P.
